摘要
选取自主培育的富硒甘薯,打磨成粉后经预处理,以脱色率、脱蛋白率和多糖损失率为考察指标,从上样量、上样体积、洗脱速率及吸附平衡时间四个方面来选择聚酰胺柱层析法脱蛋白的工艺条件为:上样量600 mg、上样体积60 mL(2/5倍柱体积)、吸附时间20 min、洗脱速率4.5 mL/min.结果显示,虽然聚酰胺柱层析法脱蛋白的效果比Sevag法略低,但其具有良好的脱色效果,对环境无污染兼具脱色功能,考虑到甘薯硒多糖可最终制成保健品,甚至是医药用品,所以聚酰胺柱层析法是良好的脱蛋白工艺.
The self-cultivated selenium-enriched sweet potato was pretreated after being ground into powder,and the decolorization rate,deproteinization rate,and polysaccharide loss rate were used as indicators for investigation.From the four aspects of sample load,sample volume,elution rate,and adsorption equilibrium time,the process conditions for deproteinization by polyamide column chromatography were selected as follows:a loading amount of 600 mg,a loading volume of 60 mL,an adsorption time of 20 min,and an elution rate of 4.5 mL/min.The results show that although the deproteinization effect of the polyamide column chromatography is slightly lower than that of the Sevag method,it has a good decolorization effect,has no pollution to the environment and has a decolorization function.Considering that sweet potato selenium polysaccharide is finally made into a health product for medical supplies,polyamide column chromatography is a good deproteinization process.
