摘要
目的建立测定广东合欢花中木兰花碱含量的超高效液相色谱-质谱联用法。方法样品粉末经甲醇溶解超声提取1h后,采用色谱柱:ACQUITY UPLCr CSHT C181.7μm(2.1mm×100mm);以甲醇-0.1%甲酸溶液(1∶99)为流动相;流速0.3mL·min-1。质谱采用电喷雾离子化(ESI)方式,正离子采集,以多离子反应监测(MRM)模式对木兰花碱进行含量测定。结果木兰花碱在10.53~1053ng·L-1线性关系良好。木兰花碱平均加样回收率为100.8%。结论该方法快速、简便、灵敏、准确,可用于广东合欢花中木兰花碱的含量测定,可为广东合欢花的研究和质量控制提供依据。
OBJECTIVE To establish a method for the determination of magnoflorine in Magnolia coco(Lour.)DC.by UPLC-MS.METHODS The sample powder was dissolved in methanol and extracted by ultrasound for 1 hour.The column was ACQUITY UPLCr CSHT C181.7 m(2.1 mm×50 mm);the mobile phase was methanol-0.1%formic acid solution(1∶99);and the flow rate was 0.3 mL·min-1.Mass spectrometry was carried out by electrospray ionization(ESI),positive ion collection,and multi ion reaction monitoring(MRM)mode.RESULTS Magnolia alkaloids had a good linear relationship between 10.53-1053 ng·L-1.The average recovery of Magnolia alkali was 100.8%.CONCLUSION The method is rapid,simple,sensitive and accurate.It can be used for the determination of magnoflorine in Magnolia coco(Lour.)DC.It can provide a basis for the research and quality control of Magnolia coco(Lour.)DC.
