DNS法测定手参肾宝胶囊中多糖含量

Determination of Polysaccharide in Shoushen Shenbao Capsules by DNS Method

目的建立一种藏药手参肾宝胶囊中多糖的测定方法,以更好地控制手参肾宝胶囊的质量。方法采用3,5-二硝基水杨酸(DNS)显色,分光光度法测定手参肾宝胶囊中多糖含量。结果样品经DNS试剂显色后,于520 nm处测定吸光度,在此波长处溶液的吸光度与葡萄糖含量呈良好线性关系,线性范围:8.597~42.984μg/ml,回归方程:A=0.01839C+0.01030,r=0.9981,平均加样回收率98.37%,RSD=1.57%(n=6)。测得3批样品,多糖含量以葡萄糖计为103.03~105.03 mg/粒。结论此方法简单、准确、可控,重现性好,可为手参肾宝胶囊质量控制提供依据。

Objective To establish a method for the determination of polysaccharide in Tibetan medicine Shoushen Shenbao to better control the quality of Shoushen Shenbao Capsules.Methods The content of polysaccharide in Shoushen Shenbao Capsules was determined by spectrophotometry after chromogenic reaction with 3,5-dinitrosalicylic acid(DNS).Results The absorbance of the sample was determined at 520 nm after chromogenic reaction with DNS.There was a good linear relationship between the absorbance of the solution and the glucose content,and the linear range was 8.597-42.984μg/ml.The regression equation was A=0.01839C+0.01030,r=0.9981.The average recovery was 98.37%,with RSD of 1.57%(n=6).The polysaccharide contents of 3 batches of samples were 103.03-105.03 mg/capsule calculated as glucose.Conclusion This method is simple,accurate,controllable and reproducible,which can provide scientific basis for quality control of Shoushen Shenbao Capsules.
