摘要 目的分析微纤丝关联蛋白2(MFAP2)在头颈部肿瘤组织中的表达,探讨其表达与头颈部肿瘤临床病理特征之间的联系。方法利用UALCAN生物信息学分析工具分析TCGA数据库中头颈部肿瘤组织中MFAP2的表达;利用RT-PCR技术分析76例头颈部肿瘤标本和正常组织中MFAP2的mRNA表达水平,并分析MFAP2的表达与临床病理特征之间的相关性。结果与44例正常组织比较,520例头颈部肿瘤组织中MFAP2基因的表达水平显著上调,即使在Ⅰ期和Ⅱ期等早期头颈部肿瘤患者中MFAP2的表达也显著上升;而且MFAP2的表达在肿瘤T1期中的表达也显著升高。不仅如此,MFAP2的表达在淋巴结未转移的头颈部肿瘤患者中的表达也显著升高。乳头瘤病毒(HPV)阳性的头颈部肿瘤患者肿瘤组织中MFAP2的表达显著低于HPV阴性的头颈部肿瘤患者。76例头颈部肿瘤组织中MFAP2 mRNA表达显著高于对应的癌旁组织,进一步分析MFAP2的表达与头颈部肿瘤患者的TNM分期、肿瘤大小、远端转移和淋巴结转移等病理特征存在相关性。结论 MFAP2在头颈部肿瘤组织中表达显著升高,并且即使在早期患者中MFAP2的表达也显著上升,并且MFAP2的表达与肿瘤大小、淋巴结转移等临床病理特征存在紧密的相关性。 Objective To investigate the expression of MFAP2 in the head and neck carcinoma, and the correlation between MFAP2 expression and the clinical pathologic features. Methods UALCAN bioinformatic portal was used to analyze the expression of MFAP2 in the TCGA head and neck carcinoma database. RT-PCR assay was applied to test the mRNA expression of MFAP2 in the 76 tumor tissue samples and corresponding normal tissues from head and neck carcinoma patients. And the correlations between MFAP2 expression and the clinical pathologic characteristics were analyzed. Results Compared with 44 cases of normal tissues, the expression of MFAP2 was significantly increased in the 520 cases of tumor tissues, even in the patients of Stage Ⅰ and Ⅱ. Moreover, MFAP2 expression was also enhanced in the patients of Grade Ⅰ. Furthermore, even in the patients without lymph node metastasis, the MFAP2 expression was remarkably increased compared with the normal tissues. The levels of MFAP2 in the head and neck carcinoma patients with HPV infection was lower than those patients without HPV infection. The mRNA levels of MFAP2 in the 76 cases of head and neck carcinoma was significantly higher than the corresponding normal tissues. The further analysis showed that the expression of MFAP2 was closely correlated with the clinical pathologic features, such as TNM stage, tumor size, distant metastasis and lymph node metastasis. Conclusion MFAP2 is overexpressed in the head and neck carcinoma, even in early stage;and the MFAP2 expression is closely correlated with the clinical pathologic characteristics, such as tumor size and lymph node metastasis.
