摘要
目的探讨肾细胞癌中斯钙素1(stanniocalcin 1,STC1)的表达水平与肾细胞癌临床预后的关系。方法采用实时定量PCR和免疫组织化学的方法检测肾细胞癌和瘤旁正常组织中STC1mRNA和蛋白的表达水平,采用Kaplan⁃Meier方法绘制生存曲线并用log⁃rank检验差异,进一步采用Cox回归分析研究肾细胞癌患者总体生存的预测因素。结果与瘤旁正常组织相比,肾细胞癌组织中STC1的mRNA和蛋白表达水平明显上调(P<0.001),STC1低表达组患者总体生存明显优于高表达组患者(P<0.001)。远处转移(P=0.009)、组织类型(P=0.013)和肿瘤坏死(P=0.017)是肾细胞癌术后总体生存的独立预测因素,而STC1表达水平不能作为独立的预后因子(P>0.05)。结论STC1在肾细胞癌中表达上调,STC1表达水平与肾细胞癌总体生存呈负相关,然而STC1表达水平不是肾细胞癌患者预后独立危险因素。
Objective To investigate the relationship between the expression of STC1 and clinical prognosis in renal cell carcinoma.Methods Real time⁃PCR and immunohistochemistry staining were respectively used to detect the mRNA and protein expression of STC1 in renal cell carcinoma and adjacent non⁃tumor tissues.Kaplan⁃Meier method was used to draw survival curve and tested by the log⁃rank test.The Cox proportional hazards regression model was applied to identify the prognosis factors for overall survival of renal cell carcinoma.Results Compared with non⁃tumor tissues,the mRNA and protein expression level of STC1 were markedly up⁃regulated in tumors(P<0.001).Only distant metastasis(P=0.009),histology type(P=0.013),and tumor necrosis(P=0.017)were independent prognostic roles for renal cell carcinoma.However,STC1 expression was failed to independently predict overall survival for renal cell carcinoma.Conclusions STC1 is obviously up⁃regulated in tumors.The STC1 expression is negatively correlated with overall survival in renal cell carcinoma,However,STC1 expression is failed to independently predict overall survival for renal cell carcinoma.
