紫草素抗雌激素对人乳腺癌MCF-7细胞周期蛋白D1表达的影响 被引量:3

摘要 目的探讨紫草素(shikonin,SK)在雌激素作用下对人乳腺癌MCF-7细胞周期蛋白(cyclin D1)表达的影响。方法体外培养人乳腺癌MCF-7细胞,不同浓度的紫草素及在雌激素干预下作用于细胞后,通过免疫组织化学技术、Western blot法检测cyclin D1蛋白的表达变化。结果与空白对照组比较,雌二醇组cyclin D1表达量显著增多,SK各组表达含量显著降低,且cyclin D1蛋白表达量随SK浓度升高逐渐被抑制(P<0.05);雌二醇干预后,SK对cyclin D1蛋白量的表达同样具有抑制作用(P<0.05)。结论SK能降低MCF-7细胞中cyclin D1蛋白水平表达,具有浓度依赖性,并且对雌激素上调cyclin D1表达具有明显的抑制作用。 Objective To investigate the effect of shikonin(SK)on the expression of cyclin D1 of MCF-7 cells in human breast cancer under the action of estrogen.Methods MCF-7 cells were cultured in vitro,and the expression of cyclin D1 was detected by immunohistochemistry and Western blot after different concentrations of shikonin and cells were treated with estrogen.Results Compared with the blank control group,the expression of cyclin D1 was significantly increased in the estradiol group,and the expression of cyclin D1 was significantly decreased in SK group.The expression of cyclin D1 was gradually inhibited with the increase of SK concentration(P<0.05).After the intervention of estradiol,the expression of cyclin D1 was also inhibited by SK(P<0.05).Conclusion SK can decrease the expression of cyclin D1 in MCF-7 cells in a concentration-dependent manner,and has a significant inhibitory effect on up-regulation of estrogen in cyclin D1 expression.
