广西产桑叶调节血脂作用及成分鉴定研究 被引量:5

摘要 目的研究广西产桑叶调节血脂作用并对调脂活性成分进行鉴定。方法采用Triton WR-1339诱导建立急性高脂血症小鼠模型,给药后分别测定小鼠血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)及低密度脂蛋白胆固醇(LDL-C)的含量,观察桑叶水提物、桑叶醇提物、桑叶水提物+桑叶醇提物的调节血脂效果,用高效液相色谱-四级杆串联飞行时间质谱(HPLC-QTOF/MS)法对调脂效果最好的一组入血成分进行鉴定。结果各组桑叶提取物均有一定的调节血脂作用,且具有剂量依赖性,高剂量组调脂效果优于低剂量组;桑叶水提物+桑叶醇提物以高剂量协同作用调脂效果最好。小鼠血清样品中检测出9个入血成分,鉴定了7个成分,其中3个原形成分,4个代谢产物,9个入血成分主要为黄酮类化合物。结论黄酮类成分是桑叶提取物发挥调脂作用的主要成分。 Objective To study the regulation effect on blood lipids and active components identification of mulberry leaves in Guangxi.Methods Animal models of hyperlipidemia induced by Triton WR-1339 were established,the serum levels including total cholesterol( TC),triglyceride( TG),high-density lipoprotein cholesterol( HDL-C) and were low-density lipoprotein cholesterol( LDL-C) were determined to observe the effect of mulberry leaf water extract,mulberry leaf alcohol extract,mulberry leaf water extract + mulberry leaf alcohol extract on blood lipid regulation. Then the chemical components in serum were identified by HPLC-Q-TOF/MS method.Results Each group of mulberry leaf extract had a certain effect on blood lipid regulation,and it was dose-dependent. The lipid-lowering effect of high-dose group was better than that of low-dose group;mulberry leaf water extract + mulberry leaf alcohol extract had the best synergistic effect on lipid-lowering at high doses. Nine blood components were detected in mouse serum samples,and seven components were identified,including three protoforms,four metabolites,and the nine blood components were mainly flavonoids. Conclusion Flavonoids are the main components of the mulberry leaf extract to exert lipid-lowering effects.
