摘要
目的 观察血液灌流(HP)对急性百草枯(PQ)中毒兔肾组织氧化应激的影响,并探讨其机制.方法 将60只普通级雄性日本大耳白兔完全随机分为四组:正常对照组(6只,0.9%氯化钠溶液50 mg/kg灌胃),空白对照组(18只,0.9%氯化钠溶液50 mg/kg灌胃后1h给予HP治疗2h),百草枯染毒组(18只,20%百草枯溶液50 mg/kg灌胃),HP治疗组(18只,20%百草枯溶液50 mg/kg灌胃后1h给予HP治疗2h).空白对照组、百草枯染毒组、HP治疗组兔分为3个观察时间亚组(第1、3、7天),每组各6只.经以上处理后,于第1、3、7天麻醉处死兔,并留取肾脏.化学比色法检测血肌酐(SCr)、尿素氮(BUN)及肾脏超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-px);ELISA检测肾脏诱导型一氧化氮合酶(iNOS)、血红素加氧酶1(HO-1).结果 与正常对照组比较,空白对照组各观察指标无变化,百草枯染毒组SCr、BUN、SOD、MDA、GSH-px、iNOS、HO-1均明显升高(均P<0.05).与百草枯染毒组比较,第1、3、7天HP治疗组SCr、BUN、SOD、MDA、GSH-px、HO-1均明显降低(均P<0.05);iNOS活力第1天差异无统计学意义(P>0.05),而第3、7天均明显降低(均P <0.05).结论 百草枯中毒可导致兔肾脏氧化/抗氧化系统失衡,进一步导致肾脏氧化损伤,对百草枯中毒所致的肾脏损伤具有保护作用.
Objective To investigate the effect of hemoperfusion(HP)on paraquat(PQ)-induced kidney oxidative stress injury of rabbit and the mechanism of it.Methods Sixty male rabbits were randomly divided into four groups,the normal control group(n=6,the rabbits were given normal saline by gavage),blank control group(n=18,he rabbits were given 2 hoursz HP once within 1 hour after given normal saline by gavage),PQ poisoning group(n=18,the rabbits were given 50 mg/kg 20%PQ solution by gavage),HP treatment group(n=18,the rabbits were given 2 hoursx HP once within 1 hour after 20%PQ solution espoused).The last three groups were divided into three observation time groups(1,3,7 day),containing 6 rabbits each group.On days 1,3 and 7,all groups rabbits were anesthetized and sacrificed,and their kidney tissues were collected.The levels of iNOS and HO-1 were measured by ELISA.The expression levels of SCr,BUN,SOD,MDA,GSH一px,were measured by chemical colorimetric method to observe oxidative stress injury.Results Compared with the normal control group rabbits,there were no changes in the SCr,BUN,SOD,MDA,GSH-px,HO-1 and iNOS of blank control group(P>0.05).Elevated levels of SCr,BUN,SOD,MDA,GSH-px,HO-1 and iNOS in the kidney tissue were caused by PQ poisoning(P<0.05),but were significantly inhibited by HP treatment(P<0.05).Activity of iNOS was not significantly changed on day 1(P>0.05),There were significant decrease on days 3,7 in HP treatment group(P<0.05).Conclusion PQ poisoning leads to kidney rabbit oxidant/antioxidant system imbalance and further causes oxidative damage.HP significantly increases resistance to PQ toxicity in the rabbit kidney and exerts a protective effect on PQ-induced kidney injury.
