摘要
目的:探讨Foxa2基因对小鼠心脏房间隔缺损的影响。方法:取NIPBL+/-基因缺陷鼠6只为实验组,NIPBL+/+基因缺陷鼠6只为对照组,检测小鼠体重观察小鼠生长发育情况,采用实时荧光定量PCR(qRT-PCR)法检测Foxa2基因表达。结果:实验组与对照组相比,小鼠的体重明显偏低,且差异具有统计学意义(P<0.05)。实验组心脏房间隔处的Foxa2基因表达明显低于对照组,且差异具有统计学意(P<0.05)。结论:NIPBL+/-基因缺陷鼠中Foxa2基因表达降低,在一定程度上导致小鼠生长发育迟缓,体重下降,可能是造成小鼠心脏房间隔缺损的原因,本实验通过探讨Foxa2基因表达对小鼠心脏房间隔缺损的影响,为先天性心脏病的诊断和干预及降低出生病患率提供了新的思路。
Objective To investigate the effect of Foxa2 gene on atrial septal defect in mice.Method 6 NIPBL+/-defective mice were used as the experimental group,and 6 NIPBL+/+defective mice as the control group.The weight of mice was measured and the growth and development of mice were observed.The expression of Foxa2 gene was detected by quantitative real-time PCR(qRT-pCR).Results Compared with the control group,the weight of mice in the experimental group was significantly lower,and the difference was statistically significant(P<0.05).Foxa2 gene expression in the atrial septum of the experimental group was significantly lower than that of the control group,and the difference was statistically significant(P<0.05).Conclusion The decreased expression of Foxa2 gene in NIPBL+/-gene deficient mice resulted in the growth retardation and weight loss of mice to some extent,which may be the cause of atrial septal defect in mice.In this study,we investigated the effect of Foxa2 gene expression on atrial septal defect in mice,providing new ideas for the diagnosis and intervention of congenital heart disease and the reduction of birth rate.
