人参茎叶总皂苷治疗2型糖尿病的网络药理学研究 被引量:13

摘要 目的采用网络药理学技术预测人参茎叶总皂苷治疗2型糖尿病(T2DM)的潜在作用靶点及机制。方法通过文献检索结合TCMSP数据库收集人参茎叶中的皂苷类成分,利用SwissTargetPrediction网站发现各成分的主要作用靶点,通过Genecards数据库筛选与T2DM相关的基因,构建成分-治疗T2DM靶点网络。通过String数据库得到治疗靶点间相互作用关系,构建疾病靶点互作网络。通过网络特征分析人参茎叶总皂苷治疗T2DM的关键作用靶点。利用DAVID在线分析网站对关键靶点进行基因富集分析及KEGG通路分析。结果研究共得到人参茎叶总皂苷成分45种,虚拟预测得到与T2DM相关靶点175个,关键靶点34个,这些靶点主要富集癌症通路、磷脂酰肌醇3-激酶/蛋白激酶B信号通路、黏附斑信号通路、ErBb信号通路等多条通路上。结论人参茎叶总皂苷通过多成分、多靶点、多通路综合发挥改善胰岛素抵抗、调节炎症反应、抗细胞凋亡等作用,为进一步防治T2DM提供科学依据。 Objective To predict the potential targets and mechanism of total ginsenosides of ginseng stems and leaves(TGGSL)to treat type 2 diabetes(T2DM)by network pharmacology.Methods The components of TGGSL were collected based on literatures retrieval combined with TCMSP database.The main targets of each component were found by using SwissTargetPrediction website,and genes associated with T2DM were screened by Genecards database,then the active components-targets network was constructed.The therapeutic targets interaction network was established through String database.The key targets of ginsenosides to treat T2DM were analyzed by network parameters.Gene enrichment analysis and KEGG pathway analysis of key targets were performed using DAVID online analysis website.Results A total of 45 ginsenosides were collected from TGGSL,175 potential targets were hypothesized as diabetes-related targets and 34 of which might be the key targets.These targets were mainly enriched cancer pathways,phosphatidylinositol 3-kinase/protein kinase B signaling pathway,focal adhesion signaling pathway,ErBb signaling pathway and other pathways.Conclusion This study reveals that TGGSL may improve insulin resistance,regulate inflammatory response,and resist apoptosis through multi-component,multi-target and multi-pathway synthesis,providing scientific basis for further prevention and treatment of T2DM.
