寿胎丸减味方对肾虚自然流产模型小鼠IL-17的影响 被引量:12

摘要 目的探讨寿胎丸减味方对肾虚自然流产(SA)模型小鼠血清白介素17(IL-17)和蜕膜组织中IL-17基因(mRNA)及蛋白表达的影响。方法CBA/J小鼠确定妊娠后,用羟基脲建立肾虚-SA模型,用寿胎丸减味方水提液高、中、低剂量进行治疗,并设立空白组。比较各组小鼠行为差异,流产率、血清IL-17和IL-17 mRNA、蛋白的表达差异。结果寿胎减味方丸干预下中药组与模型组相比,能够显著改善孕鼠的行为、流产率、血清IL-17水平;实时荧光定量逆转录聚合酶链反应(RT-qPCR)及蛋白印迹法(Western blot)结果显示:寿胎丸减味方能显著降低小鼠蜕膜组织中IL-17、IL-17 mRNA、蛋白的表达。结论寿胎丸减味方降低肾虚-SA小鼠流产率的机制,可能与降低蜕膜组织IL-17 mRNA、蛋白的表达,以及影响孕鼠血清IL-17的这一免疫调节机制有关。 Objective To investigate the effect of modified Shoutai Pills on protein expression of interleukin-17(IL-17)in serum and decidua tissue in mice model of spontaneous abortion(SA)due to kidney deficiency.Methods The kidney deficiency-SA model was established by hydroxyurea after the pregnancy of CBA/J mice.High-dose,medium-dose and low-dose of water extracts of modified Shoutai Pills were applied for treatment.The blank group was set.Behaviors,abortion rate,and the expressions of serum IL-17,IL-17 mRNA and protein in all groups were compared.Results Compared with the model group,the modified Shoutai Pills group had significant improvement in behaviors,abortion rate,serum IL-17 levels of pregnant mice.RT-qPCR and western blot showed that modified Shoutai Pills can significantly reduce the expressions of IL-17,IL-17 mRNA and protein in decidua tissue of mice.Conclusion The possible mechanism of modified Shoutai Pills in reducing kidney deficiency-SD mice abortion rate may be associated with the reduction of the expression of IL-17 mRNA and protein in decidua tissue and the immune regulation mechanism of serum IL-17.
