摘要
用鸡脑髓为研究对象,经过干燥、研磨、酸浸、过滤处理后制得样品液,运用邻二氮菲分光光度法测定鸡脑髓总铁含量.通过单因素平行实验法,分别初步测定最大吸收波长(λ)、pH、显色剂用量(v)、显色时间(t)的最佳条件.然后采用正交实验法进一步确定最优实验条件为:最大吸收波长510 nm、pH为5.25、显示剂用量1.50 mL、显色时间10 min.在此条件下,测得鸡脑髓总铁平均含量为88.65 ug/g.平均加标回收率高达99.67%,表明测定结果可信.
In this paper,chicken brain marrow was used as the research object.After drying,grinding,acid leaching and filtration,the sample solution was prepared.The total iron content of the sample was determined byo-phenanthroline spectrophotometry.Firstly,the optimum conditions of maximum absorption wavelength(λ),PH,developer dosage(v)and developering time(t)were determined by single factor parallel experiments.Then the orthogonal experiment method was used to further determine the optimum experimental conditions as follows:λ=510 nm,PH=5.25,v=1.50 mL,t=10 min.Under these conditions,the total iron content in chicken brain marrow was determined repeatedly to be 88.65 ug/g.The average recovery was 99.67%,which indicated that the result was credible.
