摘要
采用平板分离技术,从中烟100烟草品种的根际土壤中分离获得1株细菌,将该菌株编号为ZJFLH3。采用钼蓝比色法测定出ZJFLH3菌株经过62h发酵培养后上清液中的游离态磷含量明显增加,浓度达到了128.3μg/mL,据此将ZJFLH3菌株初步定性为解磷活性功能菌株。经过对ZJFLH3菌株的生长条件初步测定,结果表明:ZJFLH3菌株的适宜生长的温度范围是25~35℃,最适温度为30℃;适宜的pH值范围是6~7,最适酸碱度环境为中性偏酸;适宜生长的盐浓度范围是0.15%~0.2%,高盐不适宜菌株生长。ZJFLH3菌株可用作烟草菌肥、制剂开发的备用土著菌株。
A bacterial strain was isolated from rhizosphere soil of Chinese tobacco 100 by plate separation technique,and the strain was numbered as ZJFLH3. Molybdenum blue colorimetry was used to determine the content of free phosphorus in the supernatant of ZJFLH3 strain after 62 h fermentation culture,and the concentration reached 128.3 g/mL. Based on this, ZJFLH3 strain was preliminarily characterized as a functional strain with antiphosphorous activity. Furthermore, the growth characteristics of ZJFLH3 strain were preliminarily determined. The results showed that the suitable temperature range of ZJFLH3 strain was 25~35℃, and the optimum temperature was 30℃. The optimum pH range is 6~7,and the optimum pH environment is neutral acid. The range of salt concentration suitable for growth is 0.15%~0.2% and High salt is not suitable for strain growth. ZJFLH3 strain can be used as an alternative indigenous strain for tobacco fertilizer and preparation development.
