脑信号蛋白6D(SEMA6D)与Snail蛋白在胃癌组织高表达且与恶性临床病理指标呈正相关 被引量:3

摘要 目的研究脑信号蛋白6D(SEMA6D)和果蝇蜗牛同源物蛋白(Snail)在胃癌组织中表达及其临床病理学意义.方法选取54例胃癌患者的肿瘤组织和26例癌旁胃黏膜组织,Western blot法检测其蛋白水平,免疫组织化学染色法结合激光扫描共聚焦显微镜技术检测SEMA6D与Snail的表达和共定位现象,并分析两者的相关性及两者与患者临床病理学特征的关系.结果与癌旁胃黏膜组织比较,胃癌组织中SEMA6D与Snail的蛋白表达水平明显增高;且在胃癌组织中SEMA6D与Snail存在明显共定位现象;在分化程度越差、浸润深度越深、有淋巴结转移及TNM分期高的胃癌组织中,SEMA6D与Snail的蛋白表达量越高.结论SEMA6D与Snail在胃癌组织高表达且与胃癌的恶性临床病理指标相关. Objective To investigate the expression of semaphorin 6D(SEMA6D)and Snail and their clinicopathological implications in gastric cancer.Methods 54 cases of gastric cancer tissues and 26 paracancerous gastric mucosa were collected for detecting the expression of SEMA6D and Snail by immunohistochemistry and Western blot analysis.The co-localization of SEMA6D and Snail was observed by immunofluorescence double staining and laser scanning confocal microscopy.The correlation between SEMA6D and Snail and their relationships with the clinicopathological features of the patients were analyzed.Results Compared with the paracancerous gastric mucosa,the protein expression of SEMA6D and Snail in the gastric cancer significantly increased,and there was a significant co-localization of SEMA6D and Snail in gastric cancer.Further statistical analysis showed that the expression of SEMA6D and Snail in gastric cancer was positively correlated with the degree of differentiation,invasion,lymph node metastasis and TNM stage.Conclusion The high expression of SEMA6D and Snail in gastric cancer are related to the malignant clinicopathological indexes of gastric cancer.
