摘要 目的了解红核妇洁洗液对引起泌尿生殖道及浅表皮肤感染的常见病原菌体外抗菌作用,考察红核妇洁洗液对兔细菌性阴道炎和真菌性阴道炎的抗菌消炎治疗效果。方法①测定红核妇洁洗液对泌尿生殖道及浅表皮肤感染常见病原菌的最小抑菌浓度(MIC)和最小杀菌浓度(MBC);②测定红核妇洁洗液原液及5倍、10倍稀释液对实验菌株的杀菌曲线;③制备大耳白兔细菌性和真菌性阴道炎模型,用高、中、低剂量(分别为5、10、20倍稀释)红核妇洁洗液每日2次,每次持续5min,连续7日对兔阴道进行淋洗法治疗;④测定治疗前后阴道灌洗液中金黄色葡萄球菌、大肠埃希菌和白假丝酵母菌的活菌数量;⑤测定治疗0、3、5、7日阴道灌洗液中白细胞数,评定阴道清洁度;⑥取阴道组织进行病理学检查和评分。结果①红核妇洁洗液对23株泌尿生殖道及浅表皮肤感染常见病原菌菌株均有明显的抑制和杀菌作用,MIC范围0.008~0.016mL/mL,MBC范围0.016~0.063mL/mL。②红核妇洁洗液原液对所有实验菌株作用2min均可达到100%杀菌率,5倍稀释液作用3min对细菌菌株达到100%杀菌率,作用30min对真菌菌株达到100%杀菌率;10倍稀释液作用4~5min对所有细菌菌株达到100%杀菌率;对真菌菌株作用30min杀菌率在42%~95%之间。③细菌性阴道炎治疗组:治疗7日,红核妇洁洗液中剂量治疗组兔阴道白细胞数和阴道清洁度均显著低于模型组(t值分别为7.2549、3.3905,均P<0.05);真菌性阴道炎治疗组:治疗7日,红核妇洁洗液中剂量治疗组兔阴道白细胞数和阴道清洁度显著低于模型组(t值分别为5.2532、3.1976,均P<0.05)。④治疗后,红核妇洁洗液中剂量治疗组兔阴道灌洗液中金黄色葡萄球菌活菌数、大肠埃希菌活菌数、白假丝酵母菌活菌数均低于模型组(t值分别为22.6097、21.9404、14.3982,均P<0.05)。⑤兔阴道组织病理学评分:细菌性阴道炎治疗组低于模型组(t=6.3895,P<0.05),真菌性阴道炎治疗组显著低于模型组(t=4.5219,P<0.05)。⑥红核妇洁洗液高、中、低剂量组对细菌性阴道炎治愈率分别为83.33%、66.67%和50.00%,有效率均达100.00%;对真菌性阴道炎治愈率分别为66.67%、50.00%、50.00%,有效率分别为100.00%、100.00%和83.33%。结论红核妇洁洗液在体外可以有效地抑制杀灭引起泌尿生殖道感染和浅表皮肤感染的主要细菌和真菌,对兔感染性阴道炎具有显著的抗菌消炎治疗作用。 Objective To understand the antimicrobial effect in vitro of Honghefujie(HHFJ) lotion on common pathogenic microbes causing genitourinary infection and superficial skin infection,and observe the antibacterial and anti-inflammatory effects of HHFJ lotion on bacterial vaginitis and colpomycosis in rabbits.Methods ①Determined the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of HHFJ lotion on common pathogens of genitourinary and superficial skin infection.②The bactericidal curve of HHFJ concentrate and 5 times and 10 times diluent on the experimental strains were determined.③The bacterial vaginitis and colpomycosis models in rabbits were prepared and treated with high,medium and low dose(diluted 5/10/20 times,respectively) of HHFJ lotion twice a day for 5 minutes continuously for 7 days.④The viable count of Staphylococcus aureus,Escherichia coli and Candida albicans in vaginal lavage fluid were determined before and after treatment.⑤The number of leukocytes in vaginal lavage fluid were counted on 0,3,5 and 7 days,and vaginal cleanliness was assessed.⑥Collected vaginal tissue for pathological examination and grading.Results ①HHFJ lotion had obvious inhibition and bactericidal effects on 23 strains of common pathogenic bacteria in genitourinary tract and superficial epidermis infection,with MIC ranged from 0.008 mL/mL to 0.016 mL/mL and MBC ranged from 0.016 mL/mL to 0.063 mL/mL.②The original solution of HHFJ lotion concentrated on all experimental strain after 2 minutes,the sterilization rate all can reach 100%.The sterilization rate of 5 times dilution of HHFJ lotion for 3 minutes was 100% for all bacterial strains and 100% for fungal strains for 30 minutes.And the sterilization rate of 10 times dilution of HHFJ lotion for 5 minutes was 100% for all bacterial strains and 42%-80% for fungal strains for 30 minutes.③As for bacterial vaginitis treatment group,after 7 days of treatment,the number of leucocytes and vaginal cleanliness in medium dose group were significantly lower than those in model group(t values were 7.2549 and 3.3905,respectively,both P<0.05).As for colpomycosis treatment group,after 7 days of treatment,the number of leucocytes and vaginal cleanliness in medium dose group were significantly lower than those in model group(t values were 5.2532 and 3.1976,respectively,both P<0.05).④After treatment,the viable count of Staphylococcus aureus,Escherichia and Candida albicans in vaginal lavage fluid of rabbits in the middle dose group were all significantly lower than those in the model group(t values were 22.6097,21.9404 and 14.3982,respectively,all P<0.05).⑤In addition,the vaginal histopathological score of rabbits in bacterial vaginitis treatment group and colpomycosis treatment group were all significantly lower than those in model group(t values were 6.3895 and 4.5219,respectively,both P<0.05).⑥The cure rates of bacterial vaginitis in the high,middle and low dose groups were 83.33%,66.67% and 50%,respectively,and the effective rates were 100%.Moreover,the cure rates of colpomycosis were 66.67%,50% and 50%,respectively,while the effective rates were 100%,100% and 83.33%,respectively.Conclusion The HHFJ lotion can effectively inhibit and kill the main bacteria and fungi of genitourinary infection and superficial skin infection in vitro.It has significant anti-bacterial and anti-inflammatory effects on infectious vaginitis in rabbits.
