摘要
【目的】为阐明同一个miRNA簇基因(miR-665,miR-127-5p,miR-136-3p,miR-136-5p,miR432-5p,miR432-3p和miR433)在不同组织山羊表达特征和成肌细胞中表达水平。【方法】本研究采用qRT-PCR方法检测miRNA簇基因在山羊组织以及C2C12细胞不同生肌时间点的动态表达模式。【结果】miRNA簇基因(miR-665,miR-127-5p,miR-136-3p,miR-136-5p,miR432-5p,miR-432-3p和miR433)在不同波尔山羊组织中均广泛表达。miR-127-5p,miR-136-3p,miR432-5p,miR432-3p和miR433在山羊肌肉中高表达,miR-136-5p和miR«65在肌肉表达量低。miR-136-3p表达量随肌细胞增殖和分化逐浙升高;miR-127-5p,miR432-3p,miR432-5p,miR433和miR-136-5p表达量呈现先升高后降低的趋势;miR-665在随C2C12细胞增殖和分化表达逐浙降低。【结论】本研究结果为进一步阐明miRNA簇基因调控骨骼肌的生长发育的调控功能奠定了理论基础。
[Objective]The present paper aimed to clarify miRNA cluster genes(miR-665,miR-127-5p,miR-136-3p,miR-136-5p,miR-432-5p,miR-432-3p and miR-433)in expression characteristics of goats tissues and dynamical expression of proliferating and differentiating C2C12 Cells.[Method]qRT-PCR was used to detect the expression patterns of miRNA cluster genes in different tissues of Boer goats and different growth time points at myoblast proliferating and differentiating.[Result]miRNA cluster genes(miR-665,miR-127-5p,miR-136-3p,miR-136-5p,miR-432-5p,miR-432-3p and miR-433)were widely expressed in different tissues of Boer goats,miR-127-5p,miR-136-3p,miR-432-5p,miR-432-3p and miR-433 were highly expressed in muscle of Boer goats,miR-432-5p and miR-665 were lowly expressed in muscle.Besides,miR-136-3p were expressed increasingly during proliferation and differentiation of C2C12 cells,the expression level of miR-127-5p,miR-432-3p,miR-432-5p,miR-433 and miR-136-5p increased first and then decreased,miR-665 was expressed decreasingly in C2C12 Cells.[Conclusion]The results of this study provide a theoretical basis for further clarifying miRNA cluster genes in regulation of skeletal muscle growth and development.
