摘要 目的分析Fascin-1(FSCN1)蛋白、真核起始因子4A(eIF-4A)、组蛋白乙酰转移酶(hMOF)在卵巢癌中的表达及意义。方法选择2006年1月-2012年1月南方医科大学附属东莞人民医院妇产科手术切除的卵巢癌85例、卵巢交界性肿瘤25例、卵巢良性肿瘤25例、正常卵巢组织25例患者作为研究对象,采用显色原位杂交和免疫组化染色技术检测各组间FSCN1、eIF-4A、MOF的表达情况,并进行Pearson相关性分析。结果与正常对照组、卵巢良性肿瘤组及卵巢交界性肿瘤组比较,卵巢癌组中FSCN1和eIF-4A的表达均显著增高,而hMOF的表达显著降低(χ^2/P=10.097/0.004、12.469/0.009、6.586/0.017)。卵巢癌中FSCN1和eIF4A的表达与组织学分级、FIGO分期、淋巴结转移有关(FSCN1:χ^2/P=4.371/0.033、5.673/0.017、6.377/0.025;eIF-A4:χ^2/P=4.399/0.031、6.582/0.008、6.352/0.021);hMOF的表达与FIGO分期有关(χ^2/P=4.302/0.038)。Pearson相关性分析显示,卵巢癌中FSCN1表达与eIF-4A呈正相关(r=0.251,P=0.021),与hMOF呈负相关(r=-0.277,P=0.010)。FSCN1阳性、eIF-4A阳性、hMOF阴性表达者中位无瘤生存期(DFS)均显著低于FSCN1阴性、eIF4A阴性、hMOF阳性表达者(χ^2/P=5.473/0.000、5.267/0.000、5.018/0.000)。经过Logistic回归分析,FSCN1阳性、eIF-4A阳性、hMOF阴性、组织学分级高、FIGO分期晚、淋巴结转移是卵巢癌患者DFS的危险因素。结论 FSCN1、eIF-4A、hMOF在卵巢癌中存在异常表达的情况,可能与肿瘤的恶性演进密切相关。 Objective The expression and significance of Fascin 1(FSCN1) protein,eukaryotic initiation factor 4 A(eIF-4 A),and histone acetyltransferase(hMOF) in ovarian cancer were analyzed.Methods From January 2006 to January2012,85 cases of ovarian cancer,25 cases of borderline ovarian tumor,25 cases of benign ovarian tumor and 25 cases of normal ovarian tissue were selected as subjects.The expression of FSCN 1,eIF-4 A and hMOF in each group was detected by in situ hybridization and immunohistochemistry,and the Pearson correlation was analyzed.Results Compared with the normal control group,benign ovarian tumor group,and borderline ovarian tumor group,the expression of FSCN1 and eIF-4 A in the ovarian cancer group were significantly increased,while the expression of hMOF was significantly decreased(χ^2/P=10.097/0.004,12.469/0.009,6.586/0.017).The expression of FSCN1 and eIF-4 A in ovarian cancer is related to histological grade,FIGO stage,and lymph node metastasis(FSCN1:χ^2/P=4.371/0.033,5.673/0.017,6.377/0.025;eIF-4 A:χ^2/P=4.399/0.031,6.582/0.008,6.352/0.021).hMOF expression was related to FI GO stage(χ^2/P=4.302/0.038).Pearson correlation analysis showed that FSCN1 expression in ovarian cancer was positively correlated with eIF-4 A(r=0.251,P=0.021) and negatively correlated with hMOF(r=-0.277,P=0.010).The median tumor-free survival(DFS) of FSCN1 positive,eIF-4 A positive,and hMOF negative expression were significantly lower than those of FSCN1 negative,eIF-4 A negative,and hMOF positive expression(χ^2/P=5.473/0.000,5.267/0.000,5.018/0.000).Logistic regression analysis showed that FSCN1 positive,eIF-4 A positive,hMOF negative,histological grade,FIGO stage,and lymph node metastasis were risk factors for DFS in ovarian cancer patients.Conclusion The abnormal expression of FSCN 1,eIF-4 A and hMOF in ovarian cancer may be closely related to the malignant evolution of the tumor.
