高效液相色谱法检测化妆品中的6种抗生素和甲硝唑 被引量:7

Simultaneous Determination of Six Kinds of Antibiotics and Metronidazole in Cosmetics by High Performance Liquid Chromatography

摘要 采用高效液相色谱法同时测定不同种类化妆品中6种抗生素和甲硝唑的含量。样品用甲醇-0.1 mol/L盐酸(体积比1∶1)提取,采用Phenomenex Luna C18色谱柱(4.6 mm×250 mm×5μm)分离,以乙腈(A)和质量分数0.1%甲酸(B)为流动相系统,在268 nm波长下进行检测。与标准方法相比较,该研究中流动相以甲酸代替草酸盐,排除了溶剂峰的干扰,降低管路堵塞机率;6种抗生素和甲硝唑在进样范围内线性关系良好,相关系数r≥0.995,在水、乳、膏、霜四种不同基质中低(60%)、中(100%)、高(140%)三个梯度的加样回收率范围为96.5%~110.8%,相对标准偏差范围为0.32%~1.85%(n=9)。所建立的方法简便、准确、有效,可用于化妆品中6种抗生素和甲硝唑的筛查和测定。 HPLC method was established for simultaneous determination of 6 kinds of antibiotics(minocycline hydrochloride, oxytetracycline dihydrate, tetracycline hydrochloride, chlortetracycline hydrochloride, doxycycline hydrochloride, chloramphenicol) and metronidazole in cosmetics. Samples were extracted by methanol and 0.1 mol/L hydrochloric acid(volume ratio 1/1), then separated by Phenomenex Luna C18(4.6 mm×250 mm×5 μm) column using acetonitrile and mass 0.1% formic acid as mobile phase with gradient elution, the detection wavelength was 268 nm. The calibration curves showed good linear relationship and the correlation coefficients were above 0.995, the average recoveries spiked at 60%, 100%, 140% were in the range of 96.5% to 110.8% with RSD of 0.32% to 1.85%(n=9). The established method is simple、accurate and efficient. It can be used for simultaneous determination of 6 kinds of antibiotics and metronidazole in cosmetics.
