中药益肺活血方对慢性阻塞性肺疾病大鼠血栓前状态的影响

Effect of Chinese medical herbs Yifei Huoxue recipe on prothrombotic state of rat with chronic obstructive pulmonary disease

目的:观察中药益肺活血方对慢性阻塞性肺疾病(COPD)大鼠血栓前状态的影响。方法:采用熏烟加气道内注入脂多糖的方法制备COPD大鼠模型。将60只SD大鼠按随机数字表法分为正常组、模型组、对照组以及中药高、中、低剂量组,每组10只,共6组。对照组予磺达肝葵钠250μg/kg皮下注射,模型组予等量蒸馏水灌胃,正常组不予处理,中药低、中、高组分别予益肺活血方0.25 g/kg、0.50 g/kg、1.00 g/kg灌胃。光镜下观察各组COPD大鼠肺组织形态学的变化;双抗夹心ELISA法检测COPD大鼠血清纤维蛋白原(FIB)、肿瘤坏死因子-α(TNF-α)、白细胞介素-8 (IL-8)、血管内皮细胞生长因子(VEGR)表达情况。结果:光镜下对照组管腔狭窄减轻,管壁肺泡间质可见少量炎性细胞浸润;中药各组管腔狭窄程度明显减轻,肺泡扩张融合,管壁浸润的炎性细胞明显减少。中药各组TNF-α、IL-8、FIB、VEGR较模型组均有明显改善,差异具有统计学意义(P<0.05),其中以中药中剂量组疗效最佳。结论:益肺活血方可通过干预COPD血栓进展延缓疾病进程。

Objective:To observe the effect of Chinese medical herbs Yifei Huoxue recipe on prothrombotic state of rat with chronic obstructive pulmonary disease(COPD).Methods:The model of COPD rat was prepared by smoking and intratracheal instillation of lipopolysaccharide.60 SD rats were randomly divided into 6 groups i.e.normal group,model group,control group,low-,medium-and high-dose Chinese medical herbs groups,10 rats in each group.The control group was administered 250 ug/kg Fondaparinux sodium by hypodermic injection.The model group was administered equal volume of distilled water by gastric gavage.The normal group were not treated.The low-,medium-and high-dose Chinese medical herbs groups were administered Yifei Huoxue recipe respectively at 0.25 g/kg,0.50 g/kg,and 1.00 g/kg by gastric gavage.The change of lung histomorphology of COPD rats in each group was observed under light microscope.The expressions of FIB,TNF-α,IL-8 and VEGR in serum of COPD rats were detected by double-antibody sandwich ELISA.Results:Under light microscope:luminal stenosis was mitigated,and it could be seen that a small amount of inflammatory cells infiltrated in bronchial wall and alveolar interstitiumin the control group;in each Chinese medical herbs group,luminal stenosis was significantly mitigated,alveolar dilatation was fused,inflammatory cells of infiltrating in bronchial wall were significantly reduced.Compared with model group,the expressions of TNF-α、IL-8、FIB、VEGR of each Chinese medical herbs group were significantly improved,and the difference was statistically significant(P﹤0.05).the efficacy of medium-dose Chinese medical herbs group was best.Conclusion:Yifei Huoxue recipe can delay the progression of COPD by intervening in thrombosis.
