白细胞介素17A rs2275913基因多态性与儿童哮喘的相关性分析 被引量:2

摘要 目的探讨白细胞介素17A(interleukin-17A,IL-17A)多态位点rs2275913与儿童哮喘的相关性。方法选择2016-01/2017-06月作者医院收治的100例哮喘患儿进行研究,选取同时期在作者医院进行健康体检的100名儿童作为对照组。采集两组儿童的外周静脉血,提取外周血中单个核细胞的基因组DNA并测定其浓度,应用聚合酶链式反应-限制性内切酶片段长度多态性检测技术(polymerase chain reaction-restriction fragment length polymorphism, PCR-RFLP)对IL-17A位点rs2275913的多态性进行检测。结果两组儿童的IL-17A基因rs2275913位点的多态基因型及等位基因的分布均有明显的差异,观察组的AA基因型、等位基因A的占比明显高于对照组(P<0.01),GG基因型与等位基因G的占比均明显低于对照组(P<0.05),Logistic回归分析发现基因型为AA的个体发生哮喘的风险是基因型为GG个体的2.041倍;重度哮喘患儿基因型AA占比明显高于轻度、中度患儿(P<0.05),基因型GG占比明显低于轻度、中度患儿(P<0.05),肺功能异常的患儿基因型AA占比明显高于肺功能正常的患儿(P=0.016)。结论 IL-17A基因rs2275913位点的多态基因型及等位基因的分布与哮喘患儿的发病情况明显相关,IL-17A基因rs2275913的多态性对哮喘患儿的诊断等具有预测价值。 Objective To investigate the correlation between interleukin-17 A(IL-17 A) polymorphic site rs2275913 and asthma of childhood.Methods A total of 100 cases of children with asthma admitted to author’s hospital from January 2016 to June 2017 were selected as observation group, and 100 children who came to author’s hospital for physical examination at the same period were selected as the control group. Peripheral venous blood of two groups of children were collected, genomic DNA of mononuclear cells in peripheral blood were extracted and its concentration was determined, and the polymorphism of IL-17 A site rs2275913 was detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).Results The polymorphic genotype and allele distribution of rs2275913 IL-17 A gene were significantly different between the two groups, and the proportion of AA genotype,allele A of observation group were obviously higher than those of control group(P<0.01), the proportion of GG genotype and allele G were significantly lower than those of control group(P<0.05).Logistic regression analysis found that AA genotype individual risk of deeloping asthma was 2.041 times of GG genotype individuals.The proportion of AA genotype in children with severe asthma was significantly higher than that of mild and moderate patients(P<0.05), and the proportion of GG genotype in children with severe asthma was significantly lower than that of the mild, moderate patients(P<0.05), the proportion of genotype AA of abnormal lung function of patients was significantly higher than that of normal children(P=0.016).Conclusion The polymorphism of IL-17 A gene rs2275913 and the distribution of allele are significantly correlated with the incidence of asthma in children, and the polymorphism of IL-17 A gene rs2275913 have predictive value for the diagnosis of asthma in children.
