39个木槿品种亲缘关系SRAP分析 被引量:6

摘要 【目的】从分子水平确定39个品种木槿亲缘关系,为木槿品种鉴定、品种保护及杂交育种提供理论依据.【方法】使用SRAP法扩增39个木槿(Hibiscus syriacus)品种DNA条带,根据人工读带结果,利用UPGMA法进行聚类分析,得出了39份木槿样本的亲缘关系.[结果]从256对SRAP引物中筛选了10对引物组合,共扩增88条条带,平均每对引物组合扩增8.8条条带,其中多态性条带共72条,多态位点百分率(PPB)为81.81%.39个木槿品种等位基因数(Na)为1.1810,有效等位基因数(Ne)值为1.5970,Nei's基因多样性指数(H)为0.3328,Shannon's信息指数(I)为0.4287.在遗传距离为0.759处,39份木槿样本被分为5类.【结论】不同木槿变型及变种之间的遗传距离较远,不同木槿品种之间遗传距离相对较近,木槿亲缘关系远近与木槿表型性状之间存在一定联系. 【Objective】The purpose was to determine the genetic relationship of 39 Hibiscus syriacus at the molecular level,and to provide theoretical basis for Hibiscus syriacus variety identification,variaty protection and hybrid breeding.【Method】In this test,39 Hibiscus syriacus cultivars DNA bands were amplified by SRAP method.Based on the results of artificial reading,cluster analysis was performed using the UPGMA method,and relationship of 39 Hibiscus syriacus cultivars was obtained.【Result】In this study,10 pairs of primers were selected from 256 pairs of SRAP primers,and a total of 88 bands were amplified.On average,each primer pair amplified 8.8 bands,of which 72 were polymorphic bands.The ratio of polymorphic loci(PPB)was 81.81%.The number of alleles(Na)of 39 Hibiscus syriacus was 1.1810,the number of effective alleles(Ne)was 1.5970,the Nei's gene diversity index(H)was 0.3328,and the Shannon's information index(I)was 0.4287.At the genetic distance of 0.759,39 Hibiscus syriacus cultivars were classified into 5 categories.【Conclusion】The genetic distance between different variants and varieties is relatively far,and the genetic distance between different varieties is relatively close.However,there is some link between genetic relationship and phenotypic characteristics.
