摘要
【目的】为了明确紫娟茶自然杂交后的种子所萌发的实生苗与紫娟母株的遗传差异.【方法】采用28个ISSR引物对15份紫娟茶实生苗和紫娟原种基因组DNA进行PCR扩增,并采用紫外分光光度计法检测它们的花青素含量.[结果]16份紫娟茶共扩增出92条DNA条带,其中多态性条带72条,占78.3%,基因多样性指数(H)为0.31,Shannon信息指数(I)为0.45,遗传相似范围在0.53~0.79之间,平均为0.66;聚类分析发现,在0.66水平上16份紫娟茶样可分为3组,SS14的遗传相似水平与紫娟原种最高,其花青素含量与紫娟原种最接近.【结论】紫娟茶的实生苗在经ISSR鉴定、花青素含量测量,且遗传相似水平较高的前提下,可以用于构建种质群;后续在利用紫娟实生苗时,可使用区分度较高的ISSR引物855进行筛选.
【Objective】In order to clarify the genetic difference between the seedlings germinated by the seeds of Zijuan tea and its female parent.【Method】28 ISSR primers were used to amplified the genomic DNA of 15 seedlings of Zijuan tea and its female parent,and the anthocyanin content of the transplanting live seedlings was measured by the ultraviolet spectrophotometry.【Result】The results showed that a total of 92 bands was amplified,of which 72 were polymorphic,and the average polymorphic percentage was 78.3%,The gene diversity index(H)was 0.31,and the Shannon information index(I)was 0.45.The genetic similarity range was between 0.53 and 0.79 with the average of 0.66.16 Zijuan tea samples were divided into three major groups at 0.66 level by the cluster analysis.The highest level of genetic similarity appeared between SS14 and Zijuan,and the anthocyanin content of SS14 was the most similar to that of Zijuan.【Conclusion】After ISSR identification and anthocyanin content measurement on the premise of high genetic similarity level,seedlings of Zijuan could be used to construct germplasm population.When the seedlings of Zijuan need to be used,ISSR primer 855 with high differentiation would be recommended to screen before use.
