角蛋白18对人胃癌HGC27细胞增殖、迁移、凋亡及化疗敏感性的影响

Effect of Keratin 18 on Proliferation,Migration,Apoptosis and Chemosensitivity of Human Gastric Cancer HGC27 Cells

目的探究角蛋白18(CK18)对人胃癌HGC27细胞增殖、迁移、凋亡及化疗敏感性的影响。方法通过构建CK18敲低HGC27细胞株和使用CK18单克隆抗体两种方法抑制HGC27细胞中CK18的表达。对比细胞增殖、迁移、凋亡及化疗敏感性的变化。结果分别构建CK18低表达的HGC27细胞株和使用CK18单克隆抗体抑制HGC27细胞中CK18的表达后,细胞的增殖、迁移能力明显低于野生型HGC27细胞,凋亡率和化疗敏感性明显高于野生型HGC27细胞,差异均有统计学意义(P<0.05)。结论CK18可促进人胃癌HGC27细胞的增殖,迁移,抑制细胞凋亡,降低细胞对化疗药物的敏感性。

Objective To investigate the effects of keratin 18(CK18)on proliferation,migration,apoptosis and chemosensitivity of human gastric cancer cell line HG C27.Methods We inhibited the CK18 function of HG C27 cells by constructing CK18-knockdown HG C27 cell line and using CK18 monoclonal antibody.The proliferation,migration,apoptosis and sensitivity to chemotherapy of wild-type HGC27 cells were also detected for comparison.Results After CK18 expression was inhibited by construction of CK18-knockdown HG C27 cell line or CK18 monoclonal antibody,the proliferation and migration ability of HG C27 cells were significantly lower than that of wild-type HG C27 cells(P<0.05),and the apoptosis rate was higher than that of wild-type HG C27 cells(P<0.05).Moreover,the CK18-inhibited HG C27 cells were more sensitive to chemotherapeutic drugs than wild-type HGC27 cells(P<0.05).Conclusion CK18 can increase the proliferation and migration of human gastric cancer HGC27 cells,inhibit cell apoptosis,and reduce the sensitivity of cells to chemotherapy drugs.
