摘要
目的:探讨FOXP1和PBRM1在结肠癌组织以及癌旁组织中表达情况,分析其与结肠癌患者临床病理特征及预后的关系。方法:选取75例结肠癌患者癌组织及对应癌旁组织,采用qRT-PCR法检测FOXP1和PBRM1基因表达水平,免疫组织化学法检测FOXP1和PBRM1蛋白表达,使用Kaplan-Meier方法计算FOXP1和PBRM1表达对结肠癌患者存活率的影响,Spearman相关性分析FOXP1和PBRM1表达之间的相关性,Cox回归模型分析结肠癌预后的影响因素。结果:与癌旁组织相比,结肠癌组织中FOXP1基因表达水平上调,PBRM1基因表达水平下调,差异均具有统计学意义(P<0.01)。结肠癌组织中FOXP1和PBRM1阳性表达率分别为65.33%和41.33%,癌旁组织中FOXP1和PBRM1阳性表达率分别为24.00%和77.33%。与癌旁组织相比,结肠癌组织中FOXP1阳性表达明显上升,PBRM1阳性表达明显下降,差异具有统计学意义(P<0.05)。FOXP1和PBRM1在结肠癌组织中的表达均与患者的年龄、性别以及肿瘤大小无关(P﹥0.05),而与TNM分期、分化程度和淋巴结转移相关(P<0.05)。FOXP1阳性表达患者存活率显著低于阴性表达患者(P<0.05),PBRM1阳性表达患者存活率显著高于阴性表达患者(P<0.05),FOXP1阳性表达患者存活率明显低于PBRM1阳性表达患者(P<0.05)。Spearman相关性分析结果表明,FOXP1和PBRM1表达呈负相关(r=-0.547,P<0.05)。Cox回归模型分析结果显示,FOXP1和PBRM1均是影响结肠癌预后的独立因素。结论:FOXP1和PBRM1在结肠癌中表达异常,两者均参与结肠癌发生、发展过程,可能成为结肠癌诊治的新肿瘤标记物。
Objective:To investigate the expression of FOXP1 and PBRM1 in colon cancer tissues and paracancerous tissues,and to analyze their relationships with clinicopathological characteristics and prognosis of colon cancer.Methods:The colon cancer tissues and corresponding paracancerous tissues of 75 patients with colon cancer were selected.The expression levels of FOXP1 and PBRM1 genes were detected by qRT-PCR.The protein expression of FOXP1 and PBRM1 was detected by immunohistochemistry.The influence of FOXP1 and PBRM1 expression on survival rate of colon cancer patients was calculated by Kaplan-Meier.Correlation between FOXP1 and PBRM1 expression was analyzed by Spearman correlation analysis.Cox regression model was used to analyze the prognostic factors of colon cancer.Results:Compared with paracancerous tissues,the expression level of FOXP1 gene in colon cancer tissues was up-regulated,and the expression level of PBRM1 gene was down-regulated,with statistical significances(P<0.01).The positive expression rates of FOXP1 and PBRM1 in colon cancer tissues were 65.33%and 41.33%respectively,and the positive expression rates of FOXP1 and PBRM1 in paracancerous tissues were 24.00%and 77.33%respectively.Compared with paracancerous tissues,the positive expression of FOXP1 in colon cancer tissues increased significantly,and the positive expression of PBRM1 decreased significantly,with statistical significances(P<0.05).The expression of FOXP1 and PBRM1 was not correlated with age,gender and tumor size(P>0.05),but was correlated with TNM stage,differentiation degree and lymph node metastasis(P<0.05).The survival rate of patients with FOXP1 positive expression was significantly lower than that of patients with negative expression(P<0.05),and the survival rate of patients with PBRM1 positive expression was significantly higher than that of patients with negative expression(P<0.05).The survival rate of patients with FOXP1 positive expression was significantly lower than that of PBRM1 positive expression patients(P<0.05).Spearman correlation analysis results showed there was a negative correlation between FOXP1 and PBRM1 expression(r=-0.547,P<0.05).Cox regression model analysis showed that FOXP1 and PBRM1 were independent factors affecting the prognosis of colon cancer patients.Conclusion:FOXP1 and PBRM1 are abnormally expressed in colon cancer.Both are involved in the occurrence and development of colon cancer.They may become new tumor markers for the diagnosis and treatment of colon cancer.
