基于Wnt、Notch信号通路分析烙灸对脊髓损伤大鼠的神经修复作用 被引量:2

Effect on Lao Mmoxibustion’s with Spinal Cord Injury Rats Based on Wnt and Notch Signal Pathway

摘要 目的:基于Wnt、Notch信号通路分析烙灸对脊髓损伤后大鼠的神经修复作用。方法:将60只大鼠按照随机数表法分为假手术组、模型组、甲基强的松龙组、2 h烙灸组、12 h烙灸组,采用改良Allen s模型打击装置,进行急性脊髓损伤造模。甲基强的松龙组于损伤后0.5 h给药,首次30 mg/kg,之后5.4 mg/kg给药。2 h烙灸组、12 h烙灸组分别于损伤后2 h、12 h进行烙灸治疗。分别在造模成功后第1d、第7d、第14d、第21d进行脊髓组织取材,利用Westernblot进行蛋白表达量的检测。结果:在Wnt1、Wnt3a蛋白检测中,2 h烙灸组表达量明显低于12 h烙灸组,差异具有统计学意义(P<0.05);2 h烙灸组的Hes5、Presenilin1蛋白高于12 h烙灸组,差异具有统计学意义(P<0.05)。结论:烙灸在脊髓损伤后激活Wnt及Notch信号通路,而Wnt与Notch信号通路间相互作用,可能影响烙灸治疗后相关因子的蛋白表达量。 Objective To explore the effect of Lao moxibustion on the nerve repair in rats after spinal cord injury based on Wnt and Notch signaling pathway.Methods The sixty rats were randomly divided into sham operation group,model group,methylprednisolone group,2hours Lao moxibustion group and 12hours Lao moxibustion group.Allen s method was used to make the rats model of acute spinal cord injury.The western medicine group was given medicine aftermanufacturing model 30 minutes,30 mg/kg for the first time,then 5.4mg/kg was given.The group of manufacturing model two hours to treat Lao moxibustion,the group after manufacturing model twelve hours to treat Lao moxibustion.On the 1st,7th,14th and 21th day,the injured spinal cord tissue was taken then Using Westernblot to detect the related proteins.ResultsIn the detection of Wnt1 and Wnt3a protein,the expression of two-hour Lao moxibustion group was significantly lower than thetwelve-hour Lao moxibustion groupand the difference was statistically significant(P<0.05).The difference of Hes5 and Presenilin1 protein in the two-hour Lao moxibustion group was higher than the twelve-hour Lao moxibustion group and the difference was of statistical significance(P<0.05).Conclusion Lao moxibustion were activated the Wnt and Notch signaling pathways after the spinal cord injury,and the interaction between the Wnt and Notch signaling pathway may affect the protein expression of the related factors after the Lao moxibustion treatment.
