基于公共数据库资源探索Claudin家族基因表达在卵巢癌进展中的价值 被引量:1

摘要 目的通过分析公共数据库资源初步探索Claudin家族基因表达水平与其在卵巢癌发生、发展中的作用。方法首先利用Gene Expression Omnibus数据库获取Claudin家族基因在正常卵巢组织、早期卵巢癌组织、晚期卵巢癌组织中的表达水平,然后通过R语言分析得到在上述3种组织中两两比较表达水平均有统计学差异的基因,进一步在Kaplan Meier-plotter数据库上针对差异表达基因进行预后(OS)及复发(PFS)分析。结果在正常卵巢组织、早期卵巢癌及晚期卵巢癌之间两两比较表达水平均有统计学差异(均P<0.05)的基因为7个,分别为Claudin1、4、6、7、10、14、18基因;OS及PFS分析发现,Claudin7、14的基因表达水平对卵巢癌的PFS具有显著影响(P=0.0456、0.0247);同时,Claudin4、6、10的基因表达水平对卵巢癌的OS具有显著影响(P=0.0265、0.0296、0.0278)。结论本研究揭示了Claudin家族基因与卵巢癌的进展及预后显著相关,从而为进一步研究其具体分子机制奠定工作基础,为探索卵巢癌治疗新靶点提供科学证据。 Objective To explore the role of Claudin gene family in the progress of ovarian cancer.Methods The expression levels of Claudin gene family in ovarian tissue,early ovarian cancer and advanced ovarian cancer were obtained from Gene Expression Omnibus(GEO)database;the differentially expressed genes in normal ovarian tissue,early and advanced ovarian cancer tissues were analyzed by R language.The association of gene differential expression with overall survival(OS)and progress-free survival(PFS)was analyzed in Kaplan Meier-plotter database.Results Eight differential genes were obtained:Claudin1,4,5,6,7,10,14 and 18.The gene expression of Claudin 7,14 was significantly associated with PFS of ovarian cancer(P=0.0456,P=0.0247);gene expression of Claudin 4,6,10 was correlated with OS of ovarian cancer(P=0.0265,P=0.0296,P=0.0278).Conclusion This study reveals that some members of the Claudin gene family was associated with the progression of ovarian cancer,which provides reference for further study of the roles of Claudin gene family in the progression of ovarian cancer.
