黄芪对糖尿病肾病大鼠肾脏保护作用的探究

The Research of the Protective Value of Astragalus Membranaceus on the Kidneys of Diabetic Rats

目的:观察不同剂量黄芪对链脲佐菌素(STZ)诱导的糖尿病肾病(DN)大鼠肾脏保护作用。方法:选择50只健康雌性大鼠,随机选取10只作为正常对照组,然后将剩余40只大鼠用STZ诱导为糖尿病模型,并将其平均分为糖尿病组,黄芪低、中、高剂量组(腹腔分别注射5、15、30 g/kg)。干预12周末检测各组大鼠肾脏AngⅡ、ET-1含量,TGF-β1及PKC表达水平,大鼠尿液、血清及肾皮质中8-羟基脱氧鸟苷(8-OHdG)含量及血清、肾皮质中肾皮质丙二醛(MDA)含量。结果:与正常组相比,糖尿病组MDA及8-OHdG含量与AngⅡ、ET-1、TGF-β1及PKC水平均较高(P<0.05);与糖尿病组比较,黄芪各剂量组MDA、8-OHdG含量及AngⅡ、ET-1、TGF-β1及PKC水平均较低(P<0.05),其中以高剂量组改善最为明显(P<0.05)。结论:黄芪具有明显肾脏保护作用,可减轻糖尿病大鼠肾脏氧化应激损伤程度,且存在明显的剂量效应关系。

Objective:To investigate the protective value of Astragalus Membranaceus in the kidneys of diabetic rats induced by streptozotocin.Method:Totally 50 healthy female rats were enrolled,from which 10 rats were randomly selected as normal control group.Then the rest rats were induced to diabetic rat models by injections of streptozotocin(STZ),and the successful models were divided randomly into four groups:the diabetes mellitus group,the low dosage Astragalus Membranceus group,the middle dosage Astragalus Membranceus group and the high dosage Astragalus Membranceus group.After 12 weeks of intervention,the biochemical test was used to measure the levels of TGF-β1,PKC,AngⅡand ET-1.Other tests were done to measure the activity of superoxide dismutase the content of malondialdehyde(MDA)and 8-hydroxy-deoxyguanosine(8-OHdG)in the renal cortex.Result:The levels of TGF-β1,PKC,AngⅡ,ET-1,MDA and 8-OHdG in the diabetes mellitus group were higher than those in the control group(P<0.05).And the levels of TGF-β1,PKC,AngⅡ,ET-1,MDA and 8-OHdG in the Astragalus Membranceus treatment groups were lower than those in the diabetes mellitus group(P<0.05).The Astragalus Membranceus treatment high dose group improved most obviously than other groups(P<0.05).Conclusion:The traditional Chinese herbal medicine Astragalus Membranceus have a protective value in the kidneys of diabetic,even inhibit the oxidative stress in the kidneys of diabetic rats with an obvious dose-effect relationship.
