摘要
目的探讨核仁纺缍体相关蛋白1(NUSAP1)在肝癌中的表达及其对患者生存预后的影响。方法利用基因表达谱数据动态分析数据库(GEPIA)和卡普兰-迈尔-绘图仪(Kaplan-Meierplotter)数据库分析NUSAP1在肝癌组织和正常组织中的表达情况以及对肝癌患者总体生存预后的影响,再通过肿瘤芯片数据库(Oncomine)验证NUSAP1在正常组织与肝癌组织中的表达,最后通过肿瘤免疫评估资源数据库(TIMER)分析NUSAP1在肝癌免疫微环境中的表达情况。结果GEPIA数据库检索结果显示NUSAP1在肝癌组织中高表达且在各期中均高表达,表达水平高于正常肝组织(P<0.05);NUSAP1高表达组患者总体生存率(OS)、无复发生存期(RFS)、无进展生存期(PFS)和疾病特异生存期(DSS)显著低于低表达组(P<0.05);Oncomine数据库共388项有关NUSAP1在正常肝组织与不同肝癌组织的表达情况,其中有140项研究具有显著差异(P<0.05),6项肝癌数据研究中发现NUSAP1在肝癌组织高表达(P<0.05);NUSAP1在肝癌免疫微环境中与各类免疫细胞正相关(P<0.05),NUSAP1高表达对生存预后有不利的影响(P<0.05)。结论NUSAP1在肝癌组织中高表达,与肝癌的发生发展有关,且不利于肝癌患者的总体生存预后,可作为肝癌临床预后判断的生物标志物,为相关的靶向治疗药物提供依据。
Objective To investigate the expression of NUSAP1 in hepatocellular carcinoma and its clinical significance to the survival and prognosis of the patients.Methods Firstly,the expression of NUSAP1 in hepatocellular carcinoma and normal tissues and its impact on overall survival and prognosis of patients with hepatocellular carcinoma were analyzed using GEPIA and Kaplan-Meierplotter database.Secondly,Oncomine database was used to verify the expression of NUSAP1 in normal and hepatocellular carcinoma tissues.Finally,the expression of NUSAP1 in the immune microenvironment of liver cancer was analyzed by TIMER database.Results The results of searching GEPIA database showed that NUSAP1 was highly expressed in HCC tissues and was highly expressed in all stages,and the expression level was higher than that in normal liver tissues(P<0.05).The levels of OS,RFS,PFS and DSS in NUSAP1 high expression group were significantly lower than those in low expression group(P<0.05).There were 388 items in Oncomine database about the expression of NUSAP1 in normal liver tissue and different liver cancer tissues,Among them,140 studies had significant difference(P<0.05),and 6 studies involved high expression of NUSAP1 in hepatocellular carcinoma.NUSAP1 was positively correlated with various immune cells in the immune microenvironment of liver cancer,and had significant differences,and it had a significant impact on the survival and prognosis of patients with liver cancer.Conclusions The high expression of NUSAP1 in hepatocellular carcinoma was related to the occurrence and development of hepatocellular carcinoma,and was not conducive to the overall survival and prognosis of patients with hepatocellular carcinoma.It can be used as a biomarker for judging the clinical prognosis of hepatocellular carcinoma,and providing a basis for related targeted therapy medicine.
