摘要
目的研究非小细胞肺癌患者胰岛素样生长因子3(IGFBP-3)、P53、Ki-67蛋白的表达及临床意义。方法收集57例非小细胞肺癌(NSCLC)患者的血清和组织样本,酶联免疫吸附法(ELISA)检测健康对照者及肺癌患者血清IGFBP-3的浓度,免疫组化法检测肺癌组织和癌旁组织中P53和Ki-67蛋白的表达情况,并分析它们的表达与患者的临床病理参数的关系。结果与健康对照者相比,NSCLC患者血清中IGFBP-3的含量显著降低,当IGFBP-3取临界值1139.10 ng/mL时,ROC曲线下面积最大0.817,灵敏度为81.3%,特异度为57.7%。IGFBP-3的表达与患者的吸烟史有相关性(P<0.05),P53和Ki-67蛋白表达与患者吸烟史、淋巴结转移、分化程度显著相关(P<0.05)。NSCLC中P53与IGFBP-3表达呈负相关,P53和Ki-67的表达有相关性(P<0.05)。结论IGFBP-3、P53和Ki-67表达与NSCLC患者吸烟史密切相关,联合检测血清IGFBP-3有助于监测吸烟相关肺癌的发生、发展,为NSCLC诊疗提供实验依据。
Objective To study the expression and clinical significance of insulin-like growth factor 3(IGFBP-3),P53 and Ki-67 in NSCLC.Methods Serum and tissue samples from 57 NSCLC patients were collected.Enzyme-linked immunosorbent assay(ELISA)was used to detect the serum concentration of IGFBP-3 in healthy controls and NSCLC.Immunohistochemistry was used to detect the expression of P53 and Ki-67 protein.The relationship between their expression and clinicopathological parameters was analyzed.Results Compared with healthy controls,the levels of IGFBP-3 were significantly lower in NSCLC patients(P<0.001).When IGFBP-3 concentration was 1139.10 ng/mL,the area under the ROC curve was the largest(0.817).And the sensitivity and the specificity were 81.3%and 57.7%respectively.The expression of IGFBP-3 was associated with the smoking history of patients(P<0.05).The expression of P53 and Ki-67 protein was significantly correlated with smoking history,lymph node metastasis and differentiation(P<0.05).IGFBP-3 and P53 expression were negatively correlated.The expression of P53 and Ki-67 was positively correlated(P<0.05).Conclusions The expression of IGFBP-3,P53 and Ki-67 are closely related to the smoking history of NSCLC patients.Combined the detection of IGFBP-3,P53 and Ki67 can help monitor nicotine-related lung cancer and provide experimental evidence for NSCLC diagnosis and therapy.
