丹酚酸B对肠缺血再灌注损伤大鼠模型肠道的保护作用 被引量:3

Protective Effect of Salvianolic Acid B on Intestinal Ischemia-reperfusion Injury in Rats

摘要 目的研究丹酚酸B(SAB)对肠缺血再灌注损伤(IIRI)大鼠模型肠道的保护性效应。方法采用随机数字表法将48只健康雄性SD大鼠随机分为IIRI组、SAB+IIRI组、阴性对照组和SAB+阴性对照组4组,每组12只。按照试剂盒说明书测定大鼠回肠丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性,实时荧光定量RT-PCR法检测大鼠回肠炎症性因子的转录水平,ELISA法检测炎症因子的分泌水平,组织病理学检测观察IIRI大鼠肠道组织学病变及对肠道通透性的影响。结果IIRI组的MDA含量明显高于阴性对照组(P=0.005)和SAB+IIRI组(P=0.012),SOD活性明显低于阴性对照组(P=0.006)和SAB+IIRI组(P=0.017)。IIRI组的肿瘤坏死因子-α(TNF-α)(P=0.003,P=0.009)、白细胞介素(IL)-1β(P=0.026,P=0.005)、IL-6(P=0.015,P=0.003)和核因子κB(NF-κB)(P=0.007,P=0.015)的转录水平均明显高于阴性对照组和SAB+IIRI组。IIRI组的TNF-α(P=0.002,P=0.006)、IL-1β(P=0.002,P=0.006)、IL-6(P=0.008,P=0.002)和NF-κB(P=0.026,P=0.005)的分泌水平均明显高于阴性对照组和SAB+IIRI组。IIRI组的菊粉水平明显低于阴性对照组(P=0.015),高于SAB+IIRI组(P=0.011)。IIRI组的右旋糖酐水平明显低于阴性对照组(P=0.011)和SAB+IIRI组(P=0.012)。IIRI组的葡聚糖凝胶水平明显高于阴性对照组(P=0.031)和SAB+IIRI组(P=0.020)。SAB预处理能改善大鼠回肠组织肠黏膜的水肿、坏死和绒毛剥脱现象,SAB+阴性对照组的Chiu组织学得分明显高于阴性对照组(P=0.001),SAB+IIRI组Chiu组织学得分明显低于IIRI组(P=0.001)。结论SAB预处理能够减轻IIRI,这种保护性效应可能是通过减轻肠道的氧化应激反应和炎症反应来实现的。 Objective To investigate the protective effect of salvianolic acid B(SAB)on the intestinal tract of rats after intestinal ischemia-reperfusion injury(IIRI).Methods Forty-eight healthy male SD rats were equally randomized into IIRI group,SAB+IIRI group,sham control group,and SAB+sham control group.The malonyldialdehyde(MDA)level and superoxide dismutase(SOD)activity in the ileum were measured in each group according to the kit instructions,the transcription levels of inflammatory factors in the ileum of rats were detected by real-time fluorescence quantitative RT-PCR,the secretion level of inflammatory factors was detected by ELISA,and the effects of intestinal ischemia-reperfusion on intestinal permeability and histological lesions were measured by histopathology.Results The MDA level in IIRI group was significantly higher than those in negative control group(P=0.005)and SAB+IIRI group(P=0.012).SOD activity of IIRI group was significantly lower than those of negative control group(P=0.006)and SAB+IIRI group(P=0.017).The optical densities of tumor necrosis factor-α(TNF-α)(P=0.003,P=0.009),interleukin(IL)-1β(P=0.026,P=0.005),IL-6(P=0.015,P=0.003),and nuclear factor kappa-B(NF-κB)(P=0.007,P=0.015)in IIRI group were significantly higher than those in sham control group and SAB+IIRI group.The TNF-α(P=0.002,P=0.006),IL-1β(P=0.002,P=0.006),IL-6(P=0.008,P=0.002),and NF-κB(P=0.026,P=0.005)levels in IIRI group were significantly higher than those in sham control group and SAB+IIRI group.The inulin level in IIRI group was significantly lower than that in negative control group(P=0.015)and significantly higher than that in SAB+IIRI group(P=0.011).The dextran level in IIRI group was significantly lower than those in sham control group(P=0.011)and SAB+IIRI group(P=0.012).The dextran gel level in IIRI group was significantly higher than those in sham control group(P=0.031)and SAB+IIRI group(P=0.020).SAB pretreatment remarkably improved the edema,necrosis,and villus stripping of the intestinal mucosa in the ileum of rats.The Chiu score was significantly higher in SAB+sham control group than in sham control group(P=0.001)and was significantly lower in SAB+IIRI group than in IIRI group(P=0.001).Conclusion SAB pretreatment can alleviate IIRI in rat models,and this protective effect may be achieved by alleviating oxidative stress and inflammation in the intestinal tract.
