两种铜基金属有机多面体凝胶的抑菌性能及其机理研究

摘要 目的评价两种铜基金属有机多面体凝胶的抑菌性能,并对其抑菌机理进行研究。方法采用浊度法、琼脂扩散法和急性毒性试验方法,对两种铜基金属有机多面体凝胶的抑菌性能和凝胶的安全性进行评价,并探讨其抑菌机理。结果浊度法测定结果表明,两种铜基金属有机多面体在5μmol/mL低浓度下对大肠杆菌及金黄色葡萄球菌的抑菌率均> 90%。琼脂扩散法结果表明,两种凝胶对大肠杆菌、金黄色葡萄球菌、表皮葡萄球菌、铜绿假单胞菌及枯草芽孢杆菌均具有抑菌效果。两种凝胶在Cu-MOP含量为3%范围内有良好的毒理安全性。有机多面体凝胶主要通过破坏细胞膜和诱导活性氧产生发挥抑菌作用。结论两种铜基金属有机多面体凝胶均具有较强的抑菌效果,使用安全。 Objective To evaluate the antibacterial properties of two copper metal-organic polyhedron( Cu-MOP) hydrogels and to explore its antibacterial mechanism. Methods Turbidity measurement,agar diffusion method and acute toxicity systemic test were used to evaluate the antibacterial properties and the biological security of these two Cu-MOP hydrogels,and the antibacterial mechanism was investigated. Results The results of turbidity measurement showed that the antibacterial rates of two Cu-MOP at low concentration of 5 μmol/m L on Escherichia coli and Staphylococcus aureus were> 90%. The results of agar diffusion method showed that the two hydrogels exhibited antibacterial effects against Escherichia coli,Staphylococcus aureus,Staphylococcus epidermidis,Pseudomonas aeruginosa and Bacillus subtilis. Both hydrogels had good toxicological safety in the range of 3% Cu-MOP content. Cu-MOP hydrogels played antibacterial action mainly by destroying cell membranes and inducing reactive oxygen species. Conclusion Both Cu-MOP hydrogels have strong antibacterial effects and are safe to use.
