人牙髓干细胞、牙周膜干细胞表型及生长特性分析 被引量:2

摘要 目的探讨人牙周膜干细胞与人牙髓干细胞的表型及生长特性。方法2018年7至12月选择18~25岁因正畸拔除完整第三磨牙10颗。取牙周膜干细胞、牙髓干细胞进行培养、纯化、扩增。统计细胞生长增殖OD,STRO-1、CD29、CD44、CD73、CD90、CD146、CD105、CD34、CD45表达率,集落生成率。结果牙髓干细胞培养3 d出现快速生长,第9天后进入平台期;牙周膜干细胞在培养4 d后开始快速生长,第8天进入平台期。2组在3~12 d时,牙髓干细胞细胞增殖A值显著高于牙周膜干细胞,差异有统计学意义(P<0.05)。两种细胞STRO-1、CD29、CD44、CD73、CD90、CD146、CD105均呈高表达。2种细胞STRO-1、CD29、CD44、CD73、CD90、CD146、CD105、CD34、CD45表达率比较差异无统计学意义(P>0.05)。牙髓干细胞集落形成率显著高于牙周膜干细胞,差异有统计学意义(P<0.05)。结论人牙髓干细胞与人牙周膜干细胞STRO-1、CD29、CD44、CD73、CD90、CD146、CD105均呈高表达,符合干细胞表型;人牙髓干细胞较人牙周膜干细胞增殖更快,集落形成率更高。 Objective To investigate the phenotype and growth characteristics of human periodontal stem cells and human dental pulp stem cells.Methods A total of 10 intact third molars around 18~25 weeks which were extracted during orthodontic treatment in our hospital from July 2018 to December 2018 were collected,and the periodontal membrane stem cells and dental pulp stem cells were cultured,purified and amplified in vitro.The cell growth,proliferation,OD values,and the expression rates of cell STRO-1,CD29,CD44,CD73,CD90,CD146,CD105,CD34,CD45 and colony formation rate were observed.Results The rapid growth of dental pulp stem cells was observed on the 3d after cultured in vitro,and the stem cells reached the plateau stage on the 9th day.The periodontal membrane stem cells began to grow rapidly on the 4d after cultured in vitro,and the stem cells reached the plateau stage on the 8th day.The proliferation A value of dental pulp stem cells was significantly higher than that of periodontal membrane stem cells(P<0.05).The cell STRO-1,CD29,CD44,CD73,CD90,CD146 and CD105 are highly expressed in the two kinds of stem cells,however,there were no significant differences in the expression rates of STRO-1,CD29,CD44,CD73,CD90,CD146,CD105,CD34 and CD45 between the two groups(P>0.05).The colony formation rate of dental pulp stem cells was significantly higher than that of periodontal membrane stem cells(P<0.05).Conclusion The STRO-1,CD29,CD44,CD73,CD90,CD146,CD105 in human dental pulp stem cells and human periodontal membrane stem cells are highly expressed,which is accordence in line with the stem cell phenotype.Human dental pulp stem cells proliferate more rapidly than human periodontal membrane stem cells,with higher colony formation rate.
