摘要 目的探讨mRNA在肝癌及对照正常肝组织中的表达差异,并分析其与肝癌预后的关系。方法提取GTEx和TCGA数据库中肝癌样本的mRNA表达量及肝癌生存数据。通过WilcoxTest算法比较正常组织与肿瘤组织数据的mRNA表达量。联合差异mRNA与临床生存时间作生存分析,寻找对肝癌预后产生影响的mRNA,并通过GO、KEGG富集分析其相关机制。结果共获取334个差异表达mRNA。通过生存分析发现,CFB、FCN3、CFHR2、COLEC10等110个差异mRNA与肝癌患者预后相关。GO、KEGG富集结果提示,humoral immune response、DNA damage response、drug metabolism-other enzymes、chemical carcinogenesis等与肝癌患者生存时间相关。结论共获得110个与肝癌患者总生存相关的差异基因,其可能通过humoral immune response、DNA damage response、drug metabolism-other enzymes、chemical carcinogenesis等功能集发挥生物学作用,对mRNA在肝癌领域的研究具有借鉴价值。 Objective To investigate the difference of expression of mRNA in hepatocellular carcinoma(HCC)and normal liver tissues,and to establish the relationship between the expression of RNA and the prognosis of HCC.Methods Extracting GTEx and TCGA database sample amount of mRNA expression of liver cancer and liver cancer survival data.By comparing WilcoxTest algorithm normal tissue and tumor tissue mRNA expression quantity of data.Joint discrepancy mRNA and clinical survival time for survival analysis,looking for mRNA that affected the prognosis of liver cancer,and the related mechanism was through the GO and KEGG enrichment analysis.Results A total of 334 differentially expressed mRNA were obtained.Survival analysis showed that 110 different mRNA of CFB,FCN3,CFHR2 and COLEC10 were correlated with prognosis.GO enrichment,KEGG enrichment results suggest humoral immune response,DNA damage response,drug metabolism-other enzymes,chemical carcinogenesis and other associated with liver cancer patients survival time.Conclusion A total of 110 differentially expressed genes related to the overall survival of patients with hepatocellular carcinoma were obtained.These genes may play a biological role through such functional sets as humoral immune response,DNA damage response,Drug metabolism-other enzymes,and chemical carcinogenesis,which can be used for reference in the research of mRNA in the field of hepatocellular carcinoma.
