太子参多糖治疗大鼠糖尿病足溃疡研究 被引量:6

Study on the treatment of Diabetic Foot Ulcer with Polysaccharide from Pseudostellaria Heterophylla in Rats

摘要 目的:建立大鼠糖尿病足溃疡模型,用太子参多糖进行治疗,探讨太子参多糖治疗大鼠糖尿病足溃疡的药效。方法:通过腹腔注射链脲佐菌素(streptozotocin,STZ)制备大鼠高血糖模型,注射72 h后,连续3 d监测空腹血糖值在10~25 mmol/L为高血糖模型成功大鼠;采用足部全层皮肤切除法,制备大鼠足溃疡模型。实验大鼠分为空白组(生理盐水外涂)、模型组(生理盐水外涂)、金因肽阳性组(重组人表皮细胞生长因子外用溶液)、太子参多糖外用组(太子参多糖溶液外涂)。每天观察创面愈合情况,给药后第7 d及第14 d,监测大鼠空腹血糖,采集创面照片,以Image-pro plus获取图像数据,计算创面愈合率。结果:实验周期内,空白对照组大鼠血糖均处于正常水平[(4.43±0.52)^(4.59±0.43)],高血糖模型组大鼠血糖一直维持在较高水平[(16.94±4.09)^(23.61±3.43)]。给药后第7 d、14 d,太子参多糖外用组创面愈合率明显好于模型组(P<0.05),且与阳性组相差不大(P>0.05)。给药周期内各组大鼠足溃疡创面愈合的效果为:空白对照组>金因肽阳性组>太子参多糖外用组>模型组。结论:太子参多糖具有促进高血糖大鼠足溃疡创面愈合的药效。 Objective The model of diabetic foot ulcer in rats was established and treated with Polysaccharide fromPseudostellaria heterophyllato explore the efficacy of Polysaccharide in the treatment of diabetic foot ulcer in rats.Methods The rat hyperglycemic model is prepared by intraperitoneal injection of streptozoin(40 mg/kg).Fasting blood glucose was monitored for 3 days after 72 hours of injection,and the blood glucose value between 10-25 mmol/l was successful in hyperglycemia model.The rat model of foot ulcer was established by full-thickness skin resection of foot.The experimental rats were divided into four groups,the rats in the blank control group and model group were treated with saline.The positive group was treated with recombinant human epidermis growth factor solution.The external application group of Polysaccharide from Pseudostellaria heterophylla was coated with Polysaccharide solution of Pseudostellaria heterophylla.The wound healing was observed every day.On the 7th and 14th day after administration,fasting blood glucose was monitored,wound photos were collected,and then the image data were obtained by Image-pro plus to calculate the wound healing rate.Results In the experimental period,the blood glucose of the rats in the blank control group was at normal level[(4.43±0.52~4.59±0.43)],and the blood glucose of the rats in the model group was maintained at a higher level[(16.94±4.09)^(23.61±3.43)].On the 7th and 14th day after administration,the wound healing rate in the external use group was significantly better than that in the model group(P<0.05),and was not significantly different from that in the positive group(P>0.05).The effect of the wound healing of each group of the rats in the administration period was as follows:The blank control group>The positive group>The Pseudostellaria heterophylla polysaccharide group>The model group.Conclusion Pseudostellaria heterophylla polysaccharide has the effect of promoting the wound healing of foot ulcer in hyperglycemia rats.
