摘要
香叶草基合成酶(Geranylgeranyl bisphosphate synthase,GGPPS)与囊泡运输过程中小G蛋白的翻译后修饰密切相关。文章选择同时具有2个Loxp位点基因和GGPPS基因的Ggpps-floxed小鼠与带有脂肪组织特异性启动子aP2的Cre小鼠aP2-Cre进行杂交,以获得脂肪组织GGPPS特异性缺失的小鼠Ad Ggpps KO。经过基因鉴定和多组织的定量聚合酶链反应(polymerase chain reaction,PCR)以及Western blot确认,得到的小鼠模型的脂肪组织GGPPS缺失。
Geranylgeranyl bisphosphate synthase(GGPPS) was closely related with vesicle trafficking and post-translational modifications of small G proteins. Ggpps-floxed mice with two Loxp genes and GGPPS gene and aP2-Cre mice with adipose tissue specific promoter aP2 were selected to generate adipose tissue GGPPS-specific knockdown mice Ad Ggpps KO. The results of genotyping, quantitative polymerase chain reaction(PCR) and Western blot showed that the expression of GGPPS in adipose tissue was reduced.
