万寿菊的显微鉴别及5个主要成分的含量测定 被引量:3

摘要 目的通过显微鉴别的方法对万寿菊的根、茎、叶等显微特征进行研究,同时测定不同部位主要成分的含量,为其质量控制及合理有效的开发利用提供理论依据。方法采用显微鉴定法观察万寿菊的显微结构特征;采用YMC C18色谱柱(4.6 mm×250 mm,5μm),乙腈(B)-0.1%甲酸水溶液(A)为流动相,梯度洗脱;检测波长为257 nm;柱温为室温;流速为1.0 mL/min;供试品溶液进样量为10μL。结果5,7,3′-三羟基-3,6,4′-三甲氧基黄酮(1)、5,7,4′-三羟基-3,6-二甲氧基黄酮(2)、万寿菊苷(3)、5,3′-二羟基-3,6,4′-三甲氧基-黄酮-7-O-β-D-葡萄糖苷(4)和5-羟甲基糠醛基-甲基-丁二酸酯(5)5种化合物的线性回归方程分别为:Y=2.7957X-0.1385、Y=2.7951X-0.1163、Y=0.1740X-0.0231、Y=0.9006X-0.1242、Y=0.1815X-0.0916,相关系数为0.09992~0.09995,加样回收率在96.43%~101.17%,RSD在0.13%~1.75%。万寿菊根茎中均有此5个化学成分,且除化合物5外,其余4种化合物在茎中的含量均高于根中。化合物5在万寿菊茎部的含量最高,达2.5%。结论本研究首次对万寿菊的根、茎、叶进行有效的显微鉴定及5种重要成分进行含量测定,该方法简便、准确,重复性好,为万寿菊的质量控制提供理论依据。 Objective To provide theoretical basis for quality control and rational and effective development and utilization of Tagetes erecta L.by studying the microscopical characteristics of root,stem and leaf of marigold by means of microscopic identification,and detecting the contents of main components in different parts.Methods The microstructure features of Tagetes erecta L.were observed by microscopic identification.YMC C18 column(4.6 mm×250 mm,5μm)was used as the column,acetonitrile(B)-0.1%formic acid aqueous solution(A)was used as mobile phase of gradient elution with the detection wavelength of 257 nm and the colomn temperature was room temperature.The flow rate was 1.0 mL/min.The injection volume of the sample was 10μL.Results 5,7,3′-trihydroxyl-3,6,4′-trimethoxylflavone(1),syringic acid(2),patulitrin(3),5,3′-dihydroxyl-3,6,4′-trimethoxylflavone-7-O-β-D-glucopyranoside(4),5-hydroxymethylfurfuryl methyl succinate(5)five kinds of linear regression equations were:Y=2.7957X-0.1385,Y=2.7951X-0.1163,Y=0.1740X-0.0231,Y=0.9006X-0.1242,Y=0.1815X-0.0916.The correlation coefficient of the compounds was 0.09992-0.09995,the recovery rate of the sample was 96.43%-101.17%,and the RSD was 0.13%-1.75%.These five chemical components were found in the rhizome of Tagetes erecta L.,and the contents of the other four compounds in the stem were larger than those in the root except for the compound 5.The content of compound 5 was the highest in the stem of Tagetes erecta L.,up to 2.5%.Conclusion In this experiment,the effective microscopic identification of the roots,stems and leaves of marigold and the determination of five important components are carried out for the first time.The method is simple,accurate and reproducible,and provides a theoretical basis for the quality control of Tagetes erecta L..
