摘要
目的探讨白藜芦醇(resveratrol,RES)对ST2细胞(小鼠骨髓间充质干细胞)增殖及成骨分化的作用,以期为颞下颌关节骨关节炎的治疗提供思路。方法对第3代ST2细胞分别用0、5、10、15和20μmol/L的RES培养,48 h后采用MTT检测细胞的增殖,采用Real time RT-PCR和Western blot检测成骨特异性蛋白Runx2、Osterix的表达。结果不同浓度RES对ST2细胞无显著的促增殖作用,但RES能增强ST2细胞的成骨分化能力,且具有剂量依赖性。结论 RES可促进ST2细胞的成骨分化,可能为颞下颌关节骨关节炎的治疗提供新的思路。
Objective To investigate the effect of resveratrol( RES) on the proliferation and osteogenic differentiation of ST2 cell( bone mesenchymal stem cells of mice). Methods Third-generation ST2 cells were cultured with different concentrations of RES( 0、5、10、15 和 20 μmol/L). After 48 hours,cell proliferation was detected by MTT. After 7 days of osteogenic induction,real-time RT-PCR and western blot were used to detect Runx2 and Osterix expression at mRNA and protein levels. Results RES did not greatly influence the proliferation of ST2 cells,but it could significantly enhance the osteogenic differentiation capability of ST2 cells in a dose-dependent manner. In particular at a concentration of 20 μmol/L,RES could promote Runx2 and Osterix expression of ST2 cells. Conclusion RES can promote osteogenic differentiation of ST2 cells.
