蒙成药扎冲十三味丸对大鼠心脏的亚急性毒理学研究

Study on the Sub-acute Toxicology of Mongolian Patent Medicine Zhachong Shisan Wei Pill on Heart of Rat

目的:通过动物实验对蒙成药扎冲十三味丸进行心脏亚急性毒性研究,探讨过量服用扎冲十三味丸时是否对心脏产生毒性作用,从而指导临床用药.方法:将80只大鼠随机分为:正常组、中剂量组(0.432 g·kg^-1)、高剂量组(10.8 g·kg^-1)及生药组(10.8 g·kg^-1),进行连续灌胃给药21天,每天观察一般状态,于灌药后第21天进行心率及心电图的记录和监测,第22天腹主动脉取血后并处死,进行生化指标测定、脏器系数计算及其形态学的观察.结果:高剂量组及生药组均出现不同程度的中毒反应、心律失常、血清LDH及AST水平升高(P<0.01)、血清α-HBDH有所降低(P<0.05)、形态学观察心肌细胞出现不同程度损伤.结论:中剂量安全可靠,生药组和高剂量组均存在较大心脏毒性.

Objective:Through animal experiments to study the subacute toxicity of Mongolian patent medicine Zhachong Shisan Wei Pill,to explore whether overdose of this medicine has a toxic effect on the heart,so as to guide the clinical medication.Methods:Eighty rats were randomly divided into four groups:normal control group,Mongolian patent medicine Zhachong Shisan Wei Pill middle dose group(0.432 g·kg^-1),high-dose group(10.8 g·kg^-1)and crude drug group(10.8 g·k^g-1).The drug was continuously administered by gavage for 21 days.The rats’general states were observed,the heart rate and electrocardiogram were recorded and monitored on day 21 after administration.The rat blood was taken from the abdominal aorta and sacrificed on day 22,the biochemical indexes were detected,organ coefficients were calculated,furthermore,the morphological changes were observed.Results:There were different degrees of toxic reactions,arrhythmia,serum LDH and AST increase(P<0.01)and serumα-HBDH decrease(P<0.05)in the high-dose group and the crude drug group of Zhachong Shisan Wei Pill,and Morphologically confirmed that the heart had different degrees of damage.Conclusion:The middle dose of Zhachong Shisan Wei Pill is safe and reliable,while the high-dose and crude drug of Zhachong Shisan Wei Pill has greater cardiotoxicity.
