紫杉醇联合二甲双胍对人乳腺癌细胞MCF-7生长的抑制作用的研究

摘要 目的探讨紫杉醇联合二甲双胍对人乳腺癌细胞MCF-7生长的抑制作用。方法将人乳腺癌细胞MCF-7分为对照组、紫杉醇组、联合组。紫杉醇组采用紫杉醇预处理,联合组采用二甲双胍联合紫杉醇进行预处理。CCK-8实验检测不同药物浓度及作用时间对MCF-7细胞生长的抑制效果。划痕实验检测各组乳腺癌细胞迁移距离。Transwell实验检测各组乳腺癌细胞迁移、侵袭情况。结果CCK-8实验检测结果显示,紫杉醇组、联合组不同药物浓度和作用时间的细胞生长抑制率均明显高于对照组,差异有统计学意义(P<0.05)。联合组不同药物浓度和作用时间的细胞生长抑制率均明显高于紫杉醇组,差异有统计学意义(P<0.05)。划痕实验检测结果显示,紫杉醇组、联合组的细胞迁移距离均明显少于对照组,联合组的细胞迁移距离明显少于紫杉醇组,差异有统计学意义(P<0.05)。Transwell实验检测结果显示,紫杉醇组和联合组穿过小室膜和基底膜的细胞数量均明显少于对照组,联合组穿过小室膜和基底膜的细胞数量均明显低于紫杉醇组,差异有统计学意义(P<0.05)。结论二甲双胍能够加强紫杉醇对乳腺癌细胞MCF-7生长、迁移、侵袭的抑制作用。 Objective To explore the inhibition effect of paclitaxel combined with metformin on the growth of human breast cancer cells MCF-7. Methods Human breast cancer cells MCF-7 were divided into a control group,apaclitaxel group and a combination group.The paclitaxel group was pretreated with paclitaxel,while the combination group was pretreated with metformin combined with paclitaxel.CCK-8 assay was used to detect the inhibition effect of different drug concentrations and time on the growth of MCF-7 cells.The migration distance of breast cancer cells in each group was determined by the scratch test.Transwell assay was used to detect the migration and invasion of breast cancer cells in each group. Results CCK-8 test results showed that the cell growth inhibition rates of paclitaxel group and combination group at different drug concentration and time were significantly higher than that of the control group,and the difference was statistically significant(P <0.05).The cell growth inhibition rate of the combination group at different drug concentration and time was significantly higher than that of the paclitaxel group,and the difference was statistically significant(P <0.05).The results of scratch test showed that the cell migration distance in both the paclitaxel group and the combination group was significantly shorter than that in the control group,and the cell migration distance in the combination group was significantly shorter than that in the paclitaxel group,with statistically significant differences(P <0.05).Transwell test results showed that the number of cells passing through the compartment membrane and basement membranes in the paclitaxel group and the combination group was significantly less than that in the control group,and the number of cells passing through the compartment membrane and basement membranes in the combination group was significantly less than that in the paclitaxel group,with statistically significant differences(P <0.05). Conclusion Metformin can enhance the inhibitory effect of paclitaxel on the growth,migration and invasion of breast cancer cells MCF-7.
