摘要
本实验建立了高效液相色谱法测定乳制品中富马酸二甲酯。检测波长为280 nm,WondaSil C18色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇-缓冲溶液(55∶45),柱温40℃。氨水甲醇溶液作为提取剂、超声15 min、转速5000 r·min-1、pH值6.0条件下测得富马酸二甲酯回收率最高,检出限为0.60μg·mL-1,定量限为1.82μg·mL-1,平均回收率102.05%,精密度RSD为0.053%,该方法简单、快速、准确,适用于乳制品中富马酸二甲酯检测。
A high performance liquid chromatography(HPLC)was established for the determination of dimethyl fumarate in dairy products.The detection wavelength was 280 nm,WondaSil C18 Chromatographic column(4.6 mm×250 mm,5μm).The mobile phase was methanol-Buffer solution(55∶45),and the column temperature was 40℃.The recovery of dimethyl fumarate was the highest under the condition of ammonia water methanol solution as extractant,ultrasound for 15 min,rotating speed of 5000 r·min-1 and pH value of 6.0.The detection limit was 0.60μg·mL-1,the quantitative limit was 1.82μg·mL-1,the average recovery was 102.05%,and the precision RSD was 0.053%.The method is simple,rapid and accurate,and is suitable for the detection of dimethyl fumarate in dairy products.
