摘要
为获得南美引进黄果西番莲茎段高效组培快繁技术,对影响组织培养的关键因素,包括消毒剂及处理时间、芽诱导及生根培养培养基激素及浓度进行优化筛选。研究结果表明:经过预处理水培扦插后的外植体经过0.1%HgCL2处理15 min后,污染率最低,且未污染成活率最高;外植体在MS+6-BA 2.0 mg/L+IAA 1.0 mg/L上芽诱导率最高,为96.3%;不定芽在1/2 MS+IBA 0.5~1.0 mg/L培养基上生根效果最好,生根诱导率最高可达100%。经过试验筛选获得了最优化的消毒剂处理组合、芽诱导和生根诱导培养培养基的激素组合。
In order to develop an efficient tissue culture and rapid propagation protocol for yellow passion fruit(Passiflora edulis Sims)with the stem segment as explants introduced from South America,the key factors affecting tissue culture including disinfectant and treatment time,and hormone concentrations in shoot and root inducing mediums were optimized and selected.The results showed that the explants treated with 0.1%HgCl2 for 15 min had the lowest contamination rate and the highest uncontaminated survival rate.The shoot inducing rate of the explants on MS+6-BA 2.0 mg/L+IAA 1.0 mg/L was the highest,upto 96.3%.The adventitious shoots had the best rooting effect when cultured on the cultural medium 1/2 MS+IBA 0.5~1.0 mg/L,and their highest rooting rate was up to 100%.The optimal combination of disinfectant treatment,and the optimal hormone combinations of shoot and root inducing mediums were thus established based on the screening experiment.
