枸杞抗坏血酸合成酶基因的表达特征与含量的相关分析 被引量:1

Correlation Analysis of Expression Characteristics and Content of Ascorbic Acid Synthase Gene in Lycium barbarum L.

摘要 本研究采用紫外分光光度法和实时定量PCR法,分别测定了枸杞各组织中(叶,茎,花,青果,红果)主要抗坏血酸(ascorbic acid,As A)含量以及抗坏血酸合成过程中3个相关酶基因的表达情况,分析了As A含量与各基因表达量之间的相关性。结果表明,枸杞As A在叶和茎中含量最低,青果和红果次之且表达水平较为一致,花中含量最高。As A合成过程中3个关键酶基因协同表达且各酶基因相对表达量变化规律相似。此外,发育过程合成相关酶基因表达量与As A含量相关性分析表明,GLDH、AAO、APX 3个基因共同调控了枸杞各个组织部位中As A的合成与积累,其表达量和As A含量呈现正相关,且有协同增减的表达趋势,导致不同生长时期植物组织中As A含量存在差异。进一步说明,在生态因子调控下,枸杞As A合成关键酶基因的表达影响枸杞As A的合成与积累。 In this study,ultraviolet spectrophotometry and real-time quantitative PCR were used to determine the main ascorbic acid(AsA)content in each tissue(leaf,stem,flower,green fruit and red fruit)of Lycium barbarum as well as the expression of three related enzyme genes in the process of ascorbic acid synthesis,and the correlation between As A content and the expression of various genes was analyzed.The results showed that As A content in leaves and stems of Lycium barbarum was the lowest,followed by green fruit and red fruit,and the expression level was relatively consistent.As A content in flowers was the highest.During As A synthesis,the three key enzyme genes were synergistically expressed and the changes of relative expression levels of each enzyme gene were similar.In addition,the development of gene expression related enzymes and the correlation analysis showed that As A content GLDH,AAO,APX three common genetic regulation of the Lycium barbarum synthesis and accumulation of As A in the various tissues and parts,its expression and As A content present positive correlation,and have significant increase or decrease in the expression of trend,As A content in plant tissue result in different growth periods.Furthermore,under the regulation of ecological factors,the expression of key enzyme genes in As A synthesis of Lycium barbarum affected the synthesis and accumulation of As A.
