紫黄生肌膏对大鼠人粪污染创面愈合及炎症因子的影响

Effects of Zihuang Shengji Ointment on Wound Healing and Inflammatory Factors in Rats Contaminated with Human Feces

目的观察紫黄生肌膏对大鼠人粪污染创面愈合及炎症因子的影响,探讨紫黄生肌膏的促愈机制。方法将84只大鼠随机分成正常组、模型组、贝复济组和紫黄生肌膏组,每组21只,除正常组外,其余3组大鼠均用人粪污染创面制作肛瘘术后创面模型,观察用药后第3、7、14 d各组大鼠的创面愈合率,并采用酶联免疫吸附实验(ELISA)检测各组大鼠用药后第3、7、14d肉芽组织中TNF-α、IL-6蛋白表达情况。结果用药后第3、7、14 d,模型组的创面愈合率均小于正常组、贝复济组和紫黄生肌膏组(P<0.01),紫黄生肌膏组在用药后第7、14 d的创面愈合率大于贝复济组(P<0.01);用药后第3、7、14 d模型组创面TNF-α、IL-6的蛋白表达均高于正常组、贝复济组和紫黄生肌膏组(P<0.01),用药后第7、14 d紫黄生肌膏组创面的TNF-α、IL-6的蛋白表达均低于贝复济组,差异具有统计学意义(P<0.01)。结论紫黄生肌膏能够提高大鼠人粪污染创面的愈合率,加快创面的愈合速度,且可以下调创面TNF-α、IL-6促炎细胞因子的表达,减轻创面的炎症反应。

Objective To observe the effects of Zihuang Shengji Ointment on wound healing and inflammatory factors in rats contaminated with human feces,and to explore the healing mechanism of Zihuang Shengji Ointment.Methods 84 rats were randomly divided into normal group,model group,rb-bFGF group and Zihuang Shengji ointment group,21 rats in each group.Except the normal group,the other three groups were used to make wound models after anal fistula operation with human feces contaminated wounds.The wound healing rates of rats in each group were observed on the 3 rd,7 th and 14 th day after treatment,and the ELISA was used to detect the wound healing rates of rats in each group on the 3 rd,7 th and 14 th day after treatment.The expression of TNF-a and IL-6 in granulation tissue.Results The wound healing rate of the model group was lower than that of the normal group,the rb-bFGF group and the Zihuang Shengji ointment group(P<0.01).The wound healing rate of the Zihuang Shengji ointment group was higher than that of the rb-bFGF group on the 7 th and 14 th day after treatment(P<0.01).The expression of TNF-a and IL-6 protein in the model group was higher than that of the normal group,the rb-bFGF group and the Zihuang Shengji ointment group on the 3 rd,7 th and 14 th day after treatment(P<0.01).On the 7 th and 14 th day after treatment,the expression of TNF-αand IL-6 in the Zihuang Shengji ointment group was lower than that in the rb-bFGF group,and the differences were statistically significant(P<0.01).Conclusion Zihuang Shengji ointment can improve the healing rate of human feces contaminated wounds in rats,accelerate the healing rate of wounds,and can down-regulate the expression of TNF-αand IL-6 pro-inflammatory cytokines in the wounds,and alleviate the inflammatory reaction of the wounds.
