小麦-中间偃麦草代换系CH188分子细胞学鉴定 被引量:1

摘要 中间偃麦草在小麦遗传改良中具有重要的作用。通过对小麦-中间偃麦草代换系CH188进行农艺性状、细胞学和分子标记技术鉴定,以确定其优良农艺性状及导入小麦基因组中的外源染色体。结果表明,CH188平均穗长14.60 cm,株高70.60 cm,小穗数、千粒质量等优于主栽品种;根尖细胞的有丝分裂中期染色体数目为2n=42;基于中间偃麦草第6同源群Contig序列开发了160个STS标记,其中,8个可作为识别小麦-中间偃麦草代换系CH188中6St染色体特异标记,可以推测小麦-中间偃麦草代换系CH188是小麦的一对染色体被中间偃麦草6St染色体替代;其优良的农艺性状可能来源于中间偃麦草6St染色体。CH188具有优良的农艺性状,可能是一个小麦-中间偃麦草代换系。 Thinopyrum intermedium plays an important role in genetic improvement of wheat(Triticum aestivum L.).In this study,the Triticum aestivum-Thinopyrum intermedium substitution line CH188 was identified by agronomic characters,cytology and molecular marker techniques.The results showed that compared with the parent,CH188 had excellent and stable agronomic characters,spike length was 14.6 cm,plant height was 70.6 cm,the spikelets number and the 1000-grains quality were better than the main varieties.The number of mitotic metaphase chromosomes in root tip cells was 2n=42.Based on the Contig sequence of Thinopyrum intermedium homologous group 6,160 STS markers were developed,8 of which could be used as specific markers to identify 6St chromosomes in Triticum aestivum-Thinopyrum intermedium substitution line CH188.It could be inferred that Triticum aestivum-Thinopyrum intermedium substitution line CH188 was a pair of chromosomes of wheat replaced by 6St chromosomes of Thinopyrum intermedium.The excellent agronomic characters may come from the 6St chromosome of Thinopyrum intermedium.CH188 has excellent agronomic characters,which may be a Triticum aestivum-Thinopyrum intermedium substitution line.
