不同激素配比对梅花花粉离体萌发的影响

摘要 以绿萼梅花粉为材料,在液体培养基上进行离体培养,研究蔗糖、硼酸、氯化钙和PEG 4000不同激素配比对梅花花粉萌发的影响。结果表明,激素配比为蔗糖20 g/L+硼酸150 mg/L+氯化钙10 mg/L+PEG 400015 g/L时,花粉萌发率最高,达54.34%;蔗糖和硼酸对绿萼梅花粉萌发率有极显著影响,氯化钙和PEG 4000对绿萼梅花粉萌发率影响不显著。对高质量浓度蔗糖和硼酸培养液单因素研究结果表明,蔗糖质量浓度为40 g/L时,花粉萌发率最高;硼酸质量浓度为300 mg/L时,花粉萌发率最高。蔗糖40 g/L+硼酸300 mg/L+氯化钙10 mg/L+PEG400015 g/L激素配比可作为绿萼梅花粉萌发培养的最佳培养基。 The effects of different hormone combinations of sucrose,boric acid,calcium chloride and PEG 4000 on pollen germination of Prunus mume were studied on liquid medium with Prunus mume f.viridicalyx as material.The results showed that the highest germination rate was 54.34%with the hormone ratio sucrose 20 g/L+boric acid 150 mg/L+calcium chloride 10 mg/L+PEG 400015 g/L.Sucrose and boric acid had a significant effect on the pollen germination rate of Prunus mume f.viridicalyx,while calcium chloride and PEG 4000 had no significant effect on the pollen germination rate.Single factor of high concentration sucrose and boric acid culture medium study results showed that the highest pollen germination rate was found at sucrose concentration 40 g/L and boric acid concentration 300 mg/L.Therefore,the hormone ratio sucrose 40 g/L+boric acid 300 mg/L+calcium chloride 10 mg/L+PEG 400015 g/L can be used as the best medium for pollen germination and culture.
