摘要
目的探讨AQP2及AQP5蛋白在豚鼠膜迷路积水模型中的表达及其相关性。方法将20只豚鼠随机分为空白组(n=10)与模型组(n=10)。模型组腹腔注射醋酸去氨加压素6μg/(kg·d),注射10 d;空白组以1 ml生理盐水进行腹腔注射,注射10 d;7 d后取材。分别应用免疫组化方法、Western-blot法检测AQP2、AQP5蛋白在豚鼠耳蜗的表达情况。结果免疫组化结果与Western-blot结果均显示,模型组中AQP2蛋白表达含量高于空白组,AQP5表达含量低于空白组,组间比较,差异均有统计学意义(P <0. 05),但二者间并无相关性(P> 0. 05)。结论 AQP2与AQP5均参与了梅尼埃病的发病机制,但二者并无相关。
Objective To investigate the expression and correlation of AQP2 and AQP5 proteins in the hydrocephalic model of guinea pig. Methods Twenty guinea pigs were randomly divided into blank group( n = 10) and model group( n = 10). In the model group,6 μg/( kg·day) DAPP was injected intraperitoneally for 10 days;in the blank group,1 ml normal saline was injected intraperitoneally for 10 days;7 days later,the material was taken. The expression of AQP2 and AQP5 protein in guinea pig cochlea was detected by immunohistochemistry and Western blot. Results Immunohistochemical results and Western-blot results showed that the expression level of AQP2 protein in the model group was higher than that in the blank group,while the expression level of AQP5 was lower than that in the blank group,and the differences were statistically significant( P < 0. 05),but there was no correlation between the two groups( P > 0. 05).Conclusion Both AQP2 and AQP5 are involved in the pathogenesis of meniere’s disease,but there is no correlation between them.
