摘要
目的:观察芪豆扶正提取物对H22荷瘤小鼠的抗肿瘤作用。方法:通过小鼠右前肢腋窝皮下接种H22小鼠肝癌细胞建立78只H22荷瘤小鼠模型,并随机分为空白组、模型组、环磷酰胺阳性对照组、芪豆扶正提取物低、中、高剂量组,每组13只,连续给药,当模型组小鼠肿瘤块几何直径>1 cm(瘤重>1 g)时,终止试验。检测平均瘤重和抑瘤率,计算脾脏、胸腺和肝脏指数,观察提取物对小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、尿素氮(Ur)和肌酐(Cr)的影响,HE染色观察小鼠瘤块组织病理学变化。结果:芪豆扶正提取物高、中剂量组对荷瘤小鼠的抑瘤率分别为7.48%、31.25%,各剂量组对H22荷瘤小鼠肝脏肿大具有一定的抑制作用,对胸腺和脾脏的影响较小;提取物各剂量组H22荷瘤小鼠血清ALT活性具有升高的趋势,但差异均无统计学意义(P>0.05),而血清AST活性明显升高,差异均有统计学意义(P<0.05),血清Cr含量差异均无统计学意义(P>0.05);HE染色后中、高剂量组对小鼠组织肿瘤细胞有抑制作用。结论:芪豆扶正提取物中剂量组对KM小鼠肝癌H22移植瘤有抑制作用。
Objective:To observe anti-tumor effects of Qidou Fuzheng extracts on H22 tumor-bearing mice.Methods:All 78 H22 tumor-bearing mice models were established by hypodermic inoculation of H22 mice hepatoma carcinoma cells(HCC)at the armpit of right front limb,and they were randomized into the blank group,the model group,cyclophosphamide positive control group,low,moderate and high dosages groups of Qidou Fuzheng extracts,13 mice each group they accepted consecutive administration,the experiment was ceased when the geometric diameter of tumor in model mice was greater than 1 cm(tumor weight>1 g).To detect average tumor weight and tumor inhibition rate,calculate splenic index,thymus index and liver index,and observe the effects of the exracts on ALT,AST,Ur and Cr of the mice,HE staining was used to observe histopathological changes of mice tumor.Results:Tumor inhibition rates of high,moderate dosages groups of extract on tumor-bearing mice were7.48%,31.25%,different dosages groups possessed the certain inhibitory effects on liver enlargement of H22 tumor-bearing mice,smaller effects on thymus and spleen;ALT activity of H22 tumor-bearing mice in different dosages groups of the extracts showed the rising trend,while the difference had no statistical meaning(P>0.05),while AST activity rose notably,and the difference had statistical meaning(P<0.05),the difference had no statistical meaning in the contents of Cr(P>0.05);the results showed that moderate and high dosage group could inhibit tumor cells after HE staining of mice tissue.Conclusion:Moderate dosage group of Qidou Fuzheng extracts owns the inhibitory effects of liver cancer H22 transplanted tumor in KM mice.
