痛风性关节炎中IL-1β/Caspase-1介导的细胞焦亡的作用研究 被引量:5

摘要 目的分析白细胞介素1β(IL-1β)和半胱氨酸天冬氨酸蛋白水解酶1(Caspase-1)在痛风性关节炎中的表达情况,探讨细胞焦亡(pyroptosis)是否参与痛风性关节炎疾病的发生发展。方法分别收集40例急性痛风性关节炎患者(AG)和健康对照者(HC)的血清标本,同时提取其外周血单个核细胞(PBMCs),采用全自动生化分析仪检测血清中尿酸(UA)、肌酐(CREA)、血尿素氮(BUN)、谷氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总蛋白(TP)、白蛋白(ALB)、前白蛋白(PA)指标的含量;酶联免疫吸附法(ELISA)法检测血清中IL-1β的表达情况;免疫印迹法检测PBMCs中IL-1β和caspase-1的表达情况。结果AG组与HC组血清中TP和ALB值差异无统计学差异(P>0.05),AST、ALT差异有统计学意义(P<0.05),UA、CREA、BUN、PA差异有统计学意义(P<0.01);AG组血清中IL-1β的表达水平增高,AG组和HC组IL-1β浓度为(1.08±0.79)μg/L和(0.30±0.08)μg/L;AG组PBMCs中IL-1β和caspase-1的表达水平较HC组增加。结论急性痛风性关节炎患者血清中IL-1β和PBMCs中IL-1β、caspase-1表达都增加,细胞焦亡可能参与痛风性关节炎疾病的发生发展。 Objective To analyze the expression of interleukin-1 beta(IL-1β)and caspase-1 in gouty arthritis and explore whether pyroptosis is involved in the occurrence and development of gouty arthritis.Methods The serum samples of 40 patients with acute gouty arthritis and 40 healthy controls were collected and their peripheral blood mononuclear cells(PBMCs)were extracted.The contents of serum uric acid(UA),creatinine(CREA),blood urea nitrogen(BUN),alanine aminotransferase(ALT),aspartate aminotransferase(AST),total protein(TP),albumin(ALB),prealbumin(PA)were detected by the automated biochemical analyzer.The expression of IL-1βin serum was detected by enzyme-linked immunosorbent assay(ELISA),and the expression of IL-1βand caspase-1 in PBMCs was detected by Western blotting.Results There were no significant differences in the levels of serum TP and ALB between the acute gouty arthritis group and the healthy control group(P>0.05),while the levels of AST and ALT were statistically significant between the two groups(P<0.05).There were significant differences in the levels of UA,CREA,BUN and PA between the two groups(P<0.01).The expression level of IL-1βin serum was increased in the acute gouty arthritis group,and the concentration levels of IL-1βin the acute gouty arthritis group and healthy control group were(1.08±0.79)ng/mL and(0.30±0.08)ng/mL respectively.The expression levels of IL-1βand caspase-1 in PBMCs of the acute gouty arthritis group were increased when compared with the healthy control group.Conclusion The expression levels of IL-1βin serum and IL-1βand caspase-1 in PBMCs are increased in patients with acute gouty arthritis,and pyroptosis may be involved in the occurrence and development of gouty arthritis.
