长链非编码RNA-ATB在结直肠癌组织中表达及临床意义 被引量:5

摘要 目的探讨长链非编码RNA-ATB(LncRNA-ATB)在结直肠癌组织中表达及临床意义。方法选取2012年4月至2014年4月在我院行手术切除治疗的结直肠癌患者117例,实时荧光定量PCR术检测结直肠癌和癌旁组织中LncRNA-ATB表达,所有患者随访,截止日期为2019年4月30日,生存分析采用Kaplan-Meier法,影响生存时间的多因素分析采用Cox比例风险回归模型。结果 LncRNA-ATB在结直肠癌组织中相对表达量为(2.26±0.20),高于癌旁组织的(1.00±0.12),差异有统计学意义(t=57.756,P<0.001);LncRNA-ATB在浸润深度浆膜及浆膜外、低分化、TNM分期Ⅲ期、发生脉管浸润及淋巴结转移的结直肠癌组织中相对表达量高于浸润深度浆膜下、中高分化、TNM分期Ⅰ-Ⅱ期、未发生脉管浸润及淋巴结转移,差异有统计学意义(P<0.05);Kaplan-Meier生存分析结果显示,低表达组患者平均生存时间(62.84±5.58)个月,累积生存率68.75%,分别高于高表达组的(41.18±3.27)和32.94%,Log-Rank检验差异有统计学意义(χ2=9.028,P=0.003);Cox比例风险回归模型结果显示,脉管浸润(HR=2.137,P<0.05)、淋巴结转移(HR=2.230,P<0.05)和LncRNA-ATB表达(HR=2.208,P<0.05)均是患者预后的独立影响因素。结论结直肠癌组织中LncRNA-ATB呈高表达,且与患者恶性临床指标及不良预后有关,是影响患者预后的独立风险因素。 Objective To investigate the expression and clinical significance of long non-coding RNA-ATB(LncRNA-ATB)in colorectal cancer.Methods A total of 117 cases of patents with colorectal cancer who underwent surgical resection were selected in our hospital from April 2012 to April 2014.The expressions of LncRNA-ATB in colorectal cancer and adjacent tissues were detected by real-time fluorescent quantitative PCR.All patients were followed up,the deadline was April 30,2019.Survival analysis was performed using Kaplan-Meier method.Multivariate analysis of survival time was performed using Cox proportional hazard regression model.Results The relative expression level of LncRNA-ATB in colorectal cancer tissue was(2.26±0.20),which was higher than that in adjacent tissue,which was(1.00±0.12),the difference was statistically significant(t=57.756,P<0.001).The relative expression levels of LncRNA-ATB in colorectal cancer tissues with deep infiltration serosal and serosal,low differentiation,TNM stage Ⅲ,vascular invasion and lymph node metastasis were higher than those in infiltration deep subserosal,medium-high differentiation,TNM stageⅠ-Ⅱstage,no vascular invasion and lymph node metastasis,the differences were statistically significant(P<0.05).Kaplan-Meier survival analysis showed that the average survival time and the cumulative survival rate in the low-expression group were(62.84±5.58)months and 68.75%,respectively,which were higher than those in the high expression group,which were(41.18±3.27)months and 32.94%,respectively,the Log-Rank test was statistically significant(χ2=9.028,P=0.003).Cox proportional hazard regression model showed that the vessel infiltration(HR=2.137,P<0.05),lymph node metastasis(HR=2.230,P<0.05)and the expression of LncRNA-ATB(HR=2.208,P<0.05)were independent factors influencing patients’ prognosis.Conclusion LncRNA-ATB is highly expressed in colorectal cancer tissues and is associated with malignant clinical indicators and poor prognosis.It is an independent risk factor affecting the prognosis of patients.
