摘要
目的:建立高效液相色谱法(HPLC)同时测定大鼠灌胃阳和颗粒后血清中5-羟甲基糠醛、肉桂酸、桂皮醛、甘草苷等4种成分含量的方法。方法:12只大鼠采用随机数字表法分为阳和颗粒组和空白对照组,每组6只,制备含药血清,采用高效液相色谱仪进行含量测定,以乙腈和0.1%磷酸溶液为流动相进行梯度洗脱,紫外检测波长为240 nm,流速为1.0 mL/min,柱温为25℃,进样量为10μL。结果:5-羟甲基糠醛、肉桂酸、桂皮醛、甘草苷等4种成分的线性范围分别在0.001,05~0.010,5、0.021~0.210、0.001,15~0.011,5、0.001,25~0.012,5μg的范围内;精密度、专属性、稳定性和回收率试验均符合要求。结论:HPLC可以同时测定大鼠灌胃阳和颗粒后血清中5-羟甲基糠醛、肉桂酸、桂皮醛、甘草苷等4种成分含量。
Objective: To establish a method for determining of 5-hydroxymethyl furfural, cinnamic acid,cinnamaldehyde and liquiritin in serum of rats after intragastric administration of Yanghe granules by high performance liquid chromatography(HPLC). Methods: A total of 12 rats were divided into Yanghe granule group and blank control group by random number table method, 6 rats in each group. Drug-containing serum were prepared and analyzed by HPLC. Gradient elution was performed with acetonitrile and 0.1% phosphoric acid solution as the mobile phase. The UV detection wavelength was 240 nm, the flow rate was 1.0 mL/min, the column temperature was 25 ℃, and the injection volume was 10 μL. Results: The linear ranges of 5-hydroxymethylfurfural,cinnamic acid, cinnamaldehyde and liquiritin were 0.001,05-0.010,5, 0.021-0.210, 0.001,15-0.011,5, 0.001,25-0.012,5 μg respectively, and the precision, specificity, stability and recovery tests were all in accordance with the requirements. Conclusion: HPLC can simultaneously determine the levels of 5-hydroxymethyl furfural, cinnamic acid, cinnamaldehyde and liquiritin in serum of rats after intragastric administration of Yanghe granules.
